摘　　要：目的 探讨肽基精氨酸脱亚胺酶4 （PADI4）在非小细胞肺癌（NSCLC）组织中的表达特点及其与临床病理特征、预后的关系。方法 采用实时定量聚合酶链反应方法检测NSCLC 组织及配对癌旁组织中PADI4mRNA表达水平,免疫组织化学染色方法检测NSCLC组织及癌旁组织中PADI4蛋白的表达情况,并分析其表达与患者临床病理特征及预后的关系。结果 NSCLC 组织中PADI4的mRNA相对表达量及蛋白表达情况均高于癌旁正常组织（t =-80.384,χ2 =27.66,P 值均〈0.001）,且进一步分析表明PADI4表达水平与肿瘤TNM 分期有关（χ2=22.225,P 〈0.05）。生存分析结果显示,PADI4高表达组的生存时间显著低于低表达组（P 〈0.001）,且预后更差。Cox多因素回归分析证实PADI4表达情况可作为判定NSCLC患者预后的独立危险因素（P 〈0.05）。结论PADI4在NSCLC癌组织中高表达,且与TNM 分期相关,可作为判定预后的独立因子。Objective To investigate the expression and clinical significance of peptidylarginine deiminase 4 （PADI4） in non-small cell lung cancer （NSCLC） tissues. Methods The expressions of PADI4 mRNA and protein in NSCLC tissues and para-careinoma tissues were detected by quantitative real-time polymerase chain reaction and immunohistochemistry, respectively. The correlations between PADI4 and NSCLC clinicopathological features and prognosis were also investigated. Results The expressions of PADI4 mRNA and protein were higher in NSCLC tissues than those in paired normal lung tissues （ t = -80. 384, X2 = 27.66, all P d0. 001）. Furthermore, the expression of PADI4 was associated with TNM stage （X2= 22. 225, P %0.05）. The survival analysis showed that the survival time of patients in PADI4 high-expression group was lower than that in PADI4 low-expression group （ P 〈 0.001）, indicating the poor prognosis of patients with PADI4 high-expression. The result of Cox multivariate regression analysis showed that PADI4 expression was an independent risk factor for the prognosis of NSCLC patients （ P 〈0.05）. Conclusions PADI4 is over-expressed in NSCLC tissues, and PADI4 expression is associated with TNM stage. PADI4 expression will be an independent risk factor for the prognosis of NSCLC patients.
