摘 要:RT-PCR和RACE技术首次克隆了青岛文昌鱼超氧化物歧化酶基因的全长cDNA序列1 285 bp,该基因开放阅读框全长468 bp,编码155个氨基酸,有7个可与铜锌离子结合的位点,预测分子量大约为15 718.32 Da,理论等电点为5.60,编码的蛋白质不存在亚细胞定位的信号肽和跨膜结构域,属于胞内铜锌超氧化物歧化酶。系统进化分析表明文昌鱼的胞内铜锌超氧化物歧化酶位于脊椎动物大分支和无脊椎动物大分支的中间,并且更偏向于脊椎动物。This research cloned the full-length cDNA of copper-zinc superoxide dismutase gene from Branchiostoma belcheri tsingtauense for the first time by the technology of RT-FCR and RACE. Its full-length cDNA was 1 258 bp,including a 468 bp open reading frame(ORF) encoding 155 amino acids, which had seven Cu-Zn binding sites.The research deduced that molecular weight was 15 718.32 Da and the isoelectric point was 5.60. There was no subcellular localization of signal peptide and transmembrane domain. The encoded protein belonged to icCu/Zn-SOD. The analysis of phylogenetic tree showed that the icCu/Zn-SOD from Branchiostoma belcheri tsingtaunese was located between vertebrate and invertebrate, had closer relationship with the vertebrate.
