摘　　要：目的探讨ARID2基因在原发性肝癌(hepatocellular carcinoma,HCC)组织及细胞系中的表达水平及对HCC细胞系增殖及细胞周期的影响。方法采用免疫组化检测60例HCC患者肿瘤及癌旁组织标本ARID2的表达水平,实时定量PCR(q PCR)检测ARID2在不同分化HCC细胞系及LO2细胞中的表达;将靶向作用于ARID2的小干扰RNA(small interfering RNA,siRNA)转染Hep G2细胞;利用流式细胞术、MTT法检测HCC细胞的周期变化及增殖情况。结果 HCC组织中ARID2表达水平明显高于癌旁组织(P<0.01);同时,ARID2在不同分化HCC细胞系中的表达均显著高于LO2细胞株(P<0.05)。与对照组相比,ARID2干扰组其G1期细胞明显增多(P<0.01),S期细胞明显减少(P<0.01);增殖能力明显降低(P<0.05)。结论 ARID2可作为重要的生物标志物指导临床进行个体化肿瘤治疗和监控。Objective To investigate the expression level of ARID2 gene in hepatocellular carcinoma（ HCC） tissues and cell lines,and to explore its effect on HCC cell proliferation and cell cycle. Methods The expression of ARID2 was detected by immunohistochemistry in specimens of primary cancer and their adjacent non-cancerous tissues in 60 patients with HCC. The expression of ARID2 was detected by q PCR in different differentiation of HCC cell lines. The small interfering RNA（ siRNA） targeting ARID2 was transfected into HepG2 cells,and the cell cycle changes and proliferation were detected by flow cytometry and MTT,respectively. Results The expression level of ARID2 in HCC tissues was significantly higher than that in adjacent tissues（ P 〈0. 01）. The expression of ARID2 in HCC cell lines was significantly higher than that in LO2 cell lines（ P〈 0. 05）. After siRNA targeting ARID2 was transfected,the cells in the G_1 phase were significantly increased（ P 〈0. 01）,the S phase cells decreased significantly（ P〈 0. 01） and proliferation ability（ P〈 0. 05） of Hep G2 cells were inhibited. Conclusion ARID2 can be used as an important biomarker for HCC,and it can affect the cell cycle and proliferation by targeting ARID2.
