摘　　要：目的研究槲皮素(Quercetin)对膀胱癌细胞BIU-87侵袭和凋亡的影响。方法采用不同浓度槲皮素处理人膀胱癌细胞BIU-87,应用CCK-8法检测细胞增殖,Transwell法实验观察膀胱癌细胞BIU-87的侵袭能力,流式细胞术分析细胞凋亡率,免疫印迹法检测hIAP-2和MMP-2蛋白表达。结果槲皮素可有效抑制BIU-87细胞的增殖和侵袭能力(P<0.05),一定浓度和时间的槲皮素作用于膀胱癌细胞BIU-87后可促使其凋亡(P<0.05),且hIAP-2和MMP-2蛋白的表达量下降(P<0.05)。结论槲皮素可抑制膀胱癌细胞BIU-87的增殖和侵袭能力,并促使其凋亡,其机制可能与下调hIAP-2和MMP-2蛋白表达有关。Objective To study the effect of quercetin on invasion and apoptosis of bladder cancer cell line BIU-87. Methods Human bladder cancer cell line BIU-87 was treated with quercetin at different concentrations and time. Cell proliferation was detected by CCK-8 assay. Transwell method was used to observe the changes of invasion ability of bladder cancer cell line BIU-87, and the apoptosis rate was analyzed by flow cytometry. The difference of hIAP-2 and MMP-2 protein expression was detected by Western blot. Results Quercetin can inhibit the proliferation of BIU-87 cells and inhibit the invasion of BIU-87. Quercetin can induce apoptosis of bladder cancer cell line BIU-87, and the expression of hIAP-2 and MMP-2 protein were decreased. Conclusion Quercetin can inhibit the proliferation and inva-sion of bladder cancer cell line BIU-87 and induce apoptosis of BIU-87. The mechanism may be related to down-regulation of hIAP-2 and MMP-2 protein expression.
