摘  要:目的探讨PAK5在不同胃癌演化阶段胃黏膜组织中的表达情况。方法选取于本院进行胃镜检查的患者124例,其中非萎缩性胃炎(NAG)组患者17例,慢性萎缩性胃炎(CAG)伴完全性小肠上皮化生(CIM)组24例,CAG伴不完全性小肠上皮化生(IIM)组15例,CAG伴完全性结肠上皮化生(CCIM)组26例,CAG伴不完全性结肠上皮化生(ICIM)组11例,不典型增生(DYS)组15例以及胃癌(GC)组16例。采用免疫组化(IHC)和蛋白质印记(Western blot)检测PAK5在各组胃粘膜中的表达情况。结果 7组间PAK5表达及平均光密度差异有统计学意义(P<0.01);相关性分析结果显示,IHC PAK5表达的平均光密度与疾病演化呈正相关(P<0.01),Western blot PAK5表达与疾病演化呈正相关(P<0.01)。结论 PAK5在不同胃癌演化阶段存在表达差异,其表达的升高可能与胃癌的演化密切相关。Objective To investigate the expression of p21-activated kinase 5( PAK5) in gastric mucosa at different stages of gastric cancer. Methods A total of 124 cases of endoscopic biopsies were selected as research subjects,and the expressions of PAK5 were detected in 17 non-atrophic gastritis( NAG) tissues,24 chronic atrophic gastritis( CAG) with complete intestinal metaplasia( CIM) tissues,15 CAG with incomplete intestinal metaplasia( IIM) tissues,26 CAG with complete colonic intestinal metaplasia( CCIM) tissues,11 CAG with incomplete colonic intestinal metaplasia tissues( ICIM) tissues,15 CAG with dysplasia( DYS) tissues and 16 gastric cancer( GC) tissues by immunohistochemistry( IHC) and Western blot. Results There was statistical significance in the difference of PAK5 expressions and mean optical density among the 7 groups( P〈 0. 01); correlation analysis showed that mean absorbance of PAK5 protein expression was positively correlated with evolution of disease( P 〈0. 01),PAK5 protein expression of Western blot was positively correlated with evolution of disease( P 〈0. 01). Conclusion PAK5 expression was different in different stages of gastric cancer evolution,and the increase may be closely related to the evolution of gastric cancer.
