摘 要:核膜孔蛋白在植物生长发育中起重要作用。本研究对大豆基因组中的两个Nup98同源基因(Glyma.13g33910.1和Glyma.12G36350.1)揭示了较全面的生物信息学分析、基因克隆及表达分析。为大豆生长发育调控机制研究提供方向。一系列的在线软件分析显示,大豆Nup98启动子可能参与光反应、激素信号转导及抗性调控等途径;两个蛋白均含有保守的N端FG重复结构域和C端自酶解结构域;而大豆Nup98的互作蛋白可能与内质网囊泡形成、m RNA加工、细胞骨架组装、蛋白质合成及其核质运输、信号转导等功能相关;并且其功能与核膜孔上的Nup107-160亚复合物有关。利用Clustal W软件对不同作物中的Nup98同源蛋白进行序列比对分析发现,它们相互间具有高度的序列相似性,说明Gm Nup98基因在进化上功能很保守。实时定量PCR检测Gm Nup98在大豆不同组织器官的表达量,发现两个基因在根、茎、叶、根瘤等组织器官中均有表达,但是以其中一个为主。本研究结果说明,Gm Nup98基因在大豆进化过程中分化出两个拷贝,这两个拷贝在大豆不同植株器官中发挥着相同又有各自特异的功能。因此,通过对大豆核孔蛋白Nup98基因的充分分析,为下一步对该基因功能的深入研究提供理论基础。Nucleoporin(Nup) has important functions in plant development. To reveal the mechanism of growth and regulation of soybean growth in the Nup98 homologous genes(Glyma.13 G33910.1 and Glyma.12 G36350.1) were bioinformatically analyzed with online softwares. Also, their CDS sequences were cloned and their expression were analyzed by q RT-PCR. The bioinformatics data showed that two proteins may participate in the regulations of photoreaction, hormone signal transduction and resistance. They also contained FG repeats and a conserved autoproteolytic peptidase domain in the C-terminus. Interacting proteins of Gm Nup98 included proteins related to vesiculation on endoplasmic reticulum, m RNA processing, cytoskeleton assembling, protein synthesis, nucleo-cytoplasmic transportation, signal transduction and oxidase. Several members of Nup107-160 sub-nuclear pore complex were also embraced in the interacting proteins. The homologous proteins from different crops were analyzed by Clustal W, Gm Nup98 a and Gm Nup98 b protein sequences showed highly similarity to their homologous in different species. Therefore, nucleoporin 98 genes are conserved across plant species. The expression of Gm Nup98 a and Gm Nup98 b gene in different tissues and organs were detected by real-time PCR. The result showed that Gm Nup98 were expressed in root, stem, leaf, root nodule and other organs, but one of them was dominant. Together, these data suggest that the Gm Nup98 gene differentiated into two copies in soybean evolution proecss, and they have the same as well as special functions in different organs. Therefore, these comprehensive results provide theoretical basis for the next indepth study
