摘 要:目的:建立伯氏疟原虫青蒿素抗性系鼠疟模型。方法:采用鼠-鼠间血传、青蒿素剂量递增法对伯氏疟原虫常规株(青蒿素敏感株)进行抗性培育,通过测定伯氏疟原虫青蒿素抗性株和常规株的ED50,计算抗性指数。结果:从青蒿素起始剂量126.2 mg/kg(相当于1/2 ED50)开始,目前已培育至60代,青蒿素用药剂量达2646mg/kg。第16、24、34、42代及第53代抗性指数(RI)逐渐上升,分别为3.33、8.31、12.41、14.15、15.68。结论:建立了具有中度抗性的伯氏疟原虫青蒿素抗性系鼠疟模型。Objective: To develop Plasmodium berghei rodent malaria model resistant to artemisinin. Methods: The regular Plasmodium berghei parasites was used to infect animals and gradually raising dosages of artemisinin was used in a four-day scheme of treatment. The related information of sensitivities to artemisinin was collected and the resistance index(RI) was computed. Results: The starting dose of artemisinin was 126. 2 mg/kg,which was equivalent to 1/2 ED50 in the first generation,the dose was 2646. 2 mg/kg in the 60 th generation of parasites at present,the RI continually increased,values were 3. 33,8. 31,12. 41,14. 15 and 15. 68 in generation 16,24,34,42 and 53 respectively. Conclusion: A rodent malaria model with moderately resistant strength to artemisinin is developed.
