摘　　要：目的探讨Rab11过表达对膀胱癌细胞增殖、侵袭的影响。方法在BIU-87细胞系中转染Rab11过表达质粒,用CCK8、细胞周期检测及基质胶侵袭实验分析了Rab11对细胞增殖、细胞周期和侵袭的影响,并用Western blot及RT-PCR方法检测了细胞周期相关因子cyclin D1、cyclin E及侵袭相关因子MMP9的蛋白和m RNA的表达变化。结果Rab11过表达能促进膀胱癌细胞的增殖及侵袭,并能促进细胞周期相关因子cyclin D1、cyclin E及侵袭相关因子MMP9的蛋白和m RNA的表达。结论Rab11可能作为癌蛋白参与了膀胱癌的发病机制,成为治疗膀胱癌的靶点。Objective To investigate the effect of overexpression of Rab11 on the proliferation and invasion of bladder cancer cells.Methods Rab11 plasmid was transfected into BIU-87 cell line.The effects of Rab11 on cell proliferation,cell cycle and invasion were analyzed by CCK8 kid,cell cycle assay and Matrigel invasion test respectively.The expressions of cell cycle related factors cyclin D1,cyclin E and invasion related factors MMP9 and m RNA were detected by Western blot and RT-PCR methods.Results Rab11 can promote the proliferation and invasion of bladder cancer cells,and promote the expression of cell cycle related factors cyclin D1,cyclin E and invasion related factor MMP9 protein and mRNA.Conclusion Rab11 might be involved in the pathogenesis of bladder cancer,and betherapy target for the treatment of bladder cancer.
