摘 要:目的 进一步探讨前列腺干细胞抗原(PSCA)基因rs2294008多态性与膀胱癌易感性的关系.方法 检索并筛选数据库中2016年10月之前的有关PSCA基因rs2294008多态性与膀胱癌易感性相关的研究.严格制定纳入及排除标准,提取数据,采用NOS量表进行质量评价,应用Stata 12.0软件进行Meta分析,计算合并OR值及其95% CI,并进行发表偏倚评估分析.结果 共纳入文献8篇,包括13个病例-对照研究,涉及12349病例及24836例对照.Me-ta分析结果显示:按种族及病例来源分层分析,在5基因模型中等位基因频率模型(TvsC)[OR=1.15>1,95%CI=1.12~1.19,P=0.000 <0.001,I2=0.0%];显性模型(CT+ TTvsCC)=[OR=1.27> 1,95% CI=1.20 ~1.33,P =0.000 <0.001,I2 =0.0%],隐性(TFvsCC+ CT)=[OR=1.15> 1,95% CI=1.08 ~1.21,P =0.000 <0.001,I2 =0.0%]及杂合子(CTvsCC)模型=[OR=1.25,95%CI=1.18 ~1.32,P =0.000 <0.001,I2 =0.0%],以上模型分析结果均说明:PSCArs2294008(C>T)多态性与膀胱癌显著相关.结论 PSCArs2294008(C>T)基因多态性与膀胱癌的易感性显著相关,且与种族及病例来源无关;突变型T等位基因可能为膀胱癌的易感基因位点,PSCA有望成为膀胱癌遗传易感性的生物学标记物.Objective To further explore the association of the gene polymorphism in rs2294008 site of the prostate stem cell antigen (PSCA) with bladder cancer susceptibility.Methods The studies from databases for the association between PSCA rs2294008 polymorphisms and the susceptibility of bladder cancer were searched(published of paper from the computer retrieval to September 2016).For strict inclusion and exclusion criteria,extracted data,using NOS scale for quality evaluation.Then Meta-analysis was performed using Stata 12.0 software.The pooled OR with 95% CI were calculated to assess the association strength.Heterogeneity and publication bias were evaluated.Results A total of eight documents,including 13 case-control study involving 12 349 cases and 24 836 cases.The Meta-analysis showed:Analysis was stratified by ethnically and source of cases,allele frequency model (TvsC) [OR=1.15>1,95%CI=1.12~1.19,P=0.000<0.001,I2 =0.0%],dominant model (CT+TTvsCC) =[OR=1.27> 1,95% CI=1.20~1.33,P=0.000<0.001,I2=0.0%],recessive model(TTvsCC+CT) =[OR=1.15>1,95% CI=1.08~1.21,P=0.000<0.001,I2 =0.0%],and co-dominant (CTvsCC) model =[OR =1.25,95% CI=1.18~ 1.32,P =0.000 <0.001,I2 =0.0%].The Meta-analysis of above 4 gene models were described that PSCA rs2294008 (C > T) polymorphism was significantly associated with bladder cancer.Conclusions PSCA rs2294008 (C > T) polymorphism and bladder cancer risk is significantly related and unrelated cases of ethnically and source,and mutant T allele may be bladder cancer susceptibility loci.PSCA is expected to be bladder cancer genetic susceptibility biomarkers.
