摘  要:目的探讨δ阿片受体(delta opium receptor,DOR)在乳腺癌多药耐药性中的作用。方法乳腺癌MCF-7细胞分为耐药组和对照组,耐药组细胞采用含阿霉素的培养液培养6周,建立耐药细胞株;对照组细胞正常培养。68例乳腺癌患者,其中化疗耐药者45例、化疗敏感者23例,取患者癌旁正常组织为正常对照组,取化疗耐药者和化疗敏感者乳腺癌组织为化疗耐药组和化疗敏感组,采用反转录PCR法检测2组细胞和3组组织中DOR mRNA表达水平,采用Western blot法检测2组细胞和3组组织中DOR蛋白表达水平。采用ELISA法检测化疗耐药组和化疗敏感组P-gp转运蛋白表达水平,分析DOR与P-gp转运蛋白表达的相关性。结果耐药组细胞DOR mRNA和蛋白表达水平[(180.4±9.4)%、(157.2±9.3)%]高于对照组[(100.0±7.2)%、(100.0±10.1)%](P<0.05);化疗耐药组组织中DOR mRNA和蛋白表达水平[(269.6±15.8)%、(276.8±14.9)%]高于化疗敏感组[(187.5±12.3)%、(164.6±13.5)%]和正常对照组[(100.0±6.1)%、(100.0±10.5)%](P<0.05),化疗敏感组高于正常对照组(P<0.05);化疗敏感组DOR和P-gp转运蛋白高表达率(42.2%、24.4%)均明显低于化疗耐药组(78.3%、82.6%)(P<0.05),化疗敏感组和化疗耐药组DOR高表达者P-gp转运蛋白高表达率(73.3%)高于DOR低表达者(26.7%)(P<0.05);化疗耐药组和化疗敏感组DOR蛋白表达水平与P-gp转运蛋白表达水平呈正相关(r=0.856,P=0.030)。结论 DOR的表达与乳腺癌化疗耐药具有明显相关性,有可能成为重要的化疗耐药预测指标。Objective To explore the effect of delta opium receptor (DOR) on drug resistance of breast cancer chemotherapy. Methods The breast cancer MCF-7 ceils were divided into drug-resistant group and control group. The drug-resistant cells were cultured for δ weeks in adriamycin-containing medium to establish drug-resistant cell lines. The ceils in control group were cultured normally. In 68 breast cancer patients, 45 patients were resistant to chemotherapy and 23 patients were sensitive to chemotherapy. The normal adjacent tissue was as control (normal control group). The tissue resistant to chemotherapy was as chemotherapy-resistant group and sensitive to chemotherapy was as chemotherapy-sensitive group. The expression of DOR mRNA in all groups was detected by RT-PCR method. The expression of DOR protein was detected in all groups by Western blot. The expression of P-gp transporter protein was detected in chemotherapy-resistant group and chemotherapy-sensitive group by ELISA method to analyze the correlation between DOR and P-gp transporter protein. Results The expressions of DOR mRNA and protein were significantly higher in drug-resistant group ((180. 4-±9. 4)%, (157. 2±9. 3)%) than those in control group ((100. 0±7. 2)%, (100.0±10.1)%) (P〈0.05), in chemotherapy-resistant group ((269.6±15.8)%, (276.8±14.9)%) than those in chemotherapy-sensitive group ((187. 5±12. 3)%, (164. 6±13. 5)%) and normal control group ((100. 0±6. 1)%, (100.0±10.5) %) (P〈0.05), and in chemotherapy-sensitive group than those in normal control group (P〈0.05). The high expression rates of DOR and P gp transporter protein were significantly lower in chemotherapy-sensitive group (42.2%, 24.4%) than those in chemotherapy-resistant group (78.3%, 82.6%) (P〈0.05).
