摘 要:目的研究水通道蛋白8(AQP8)在胰腺导管腺癌癌组织(PC)、癌旁正常胰腺组织(Non-PC)中的表达及其临床意义。方法通过免疫组织化学(IHC)方法检测AQP8在87例胰腺导管腺癌患者配对的石蜡包埋PC、Non-PC中的表达,并对AQP8的表达与胰腺导管腺癌患者临床资料间的关系进行分析。同时利用实时荧光定量PCR(qRT-PCR)及Western blot方法各检测10例配对新鲜冻存PC、Non-PC中AQP8的表达。结果 IHC显示AQP8主要表达于PC导管细胞的细胞质中,AQP8在PC组中的阳性表达率为79.31%(69/87),而Non-PC组阳性表达率为18.39%(16/87),两组阳性表达差异有统计学意义(P<0.001),AQP8的表达与肿瘤瘤体直径(P<0.001)、分化程度(P<0.001)、淋巴结的转移(P<0.001)以及TNM分期(P<0.001)有显著相关性。qRT-PCR及Western blot显示PC组中AQP8 mRNA及其蛋白表达量均显著高于配对的Non-PC组,两组间差异有统计学意义(P<0.01,P<0.001)。结论 AQP8的表达与胰腺导管腺癌的瘤体直径、分化程度、淋巴结的转移以及TNM分期均有显著相关性,AQP8的检测有助于胰腺导管腺癌患者病情严重程度评估。Objective To study the expression and clinical significance of the aquaporins duct adenocarcinoma. Methods paired paraffin-embedded tissues Immunohistochemistry (IHC) was used to detect the including pancreatic duct adenocarcinoma tissue (PC) an 8 ( AQP8 ) in pancreatic expression of AQP8 in d the non-carcinoma adjacent tissues of pancreatic carcinoma(Non-PC) from 87 patient with pancreatic duct adenocarcinoma and to investigate the relationship between the clinical and pathological features of pancreatic duct adenocarcinoma with AQPS. Meanwhile, real-time quantitative PCR(qRT-PCR) and Western blot were used to detect the expression of AQP8 mRNA and the protein in 10 paired frozen specimens. Results AQP8 was mainly expressed in the cytoplasm of the ductal cells in PC. The positive expression rate of AQP8 protein was significantly higher in PC 79.31% (69/87) than in Non-PC 18.39% (16/87) by immunohistochemical method, difference was considered statistically signifi- cant (P 〈 0. 001 ). The high expression of AQP8 was correlated with pancreatic tumor size( P 〈 0. 001 ), differentiation(P 〈0. 001 ), lymph node metastasis (P 〈0. 001 ) and TNM stage(P 〈0. 001). qRT-PCR and Western blot indicated that AQP8 mRNA and AQP8 were significantly upregulated in PC than in Non-PC ( P 〈 0. 01, P 〈 0. 001 ). Conclusion The expression of AQP8 is correlated with pancreatic tumor size, differentiation, lymph node metastasis and TNM stage. The detection of AQP8 helps to assess the clinical condition of pancreatic cancer.
