摘 要:目的探讨妊娠期甲减补充甲状腺激素对仔鼠行为学及Reelin蛋白表达的影响。方法雌性大鼠48只,交配成功后,随机分为正常组、甲状腺激素组和模型组各16只,正常组普通饲料喂养至仔鼠出生(PN0);另外两组在GD6-GD14期间给予丙基硫氧嘧啶,甲状腺激素组在GD15至仔鼠出生前补充甲状腺激素(L-T4),模型组普通饲料喂养至仔鼠出生,每组40只仔鼠。测量孕鼠GD6和GD15、仔鼠PN0和PN7体重,检测孕鼠血清甲状腺激素水平,进行仔鼠定位航行实验、跳台实验,测定Reelin蛋白含量。结果正常组孕鼠TT4明显高于、TSH明显低于甲状腺激素组和模型组,差异有统计学意义P<0.05,说明造模成功。GD15甲状腺激素组、模型组孕鼠体重均明显低于正常组,PN0、PN7模型组仔鼠体重明显低于正常组、甲状腺激素组,差异有统计学意义P<0.05。第一天、第二天、第三天,模型组仔鼠定位航行潜伏期、跳台实验潜伏期比正常组、甲状腺激素组明显延长,差异有统计学意义P<0.05。PN7,正常组Reelin蛋白(64.23±7.58)与甲状腺激素组(62.38±7.62)比较,差异无明显统计学意义P>0.05,模型组(28.46±9.33)明显低于正常组和甲状腺激素组,差异有统计学意义P<0.05。模型组细胞层次紊乱、稀疏、不整齐,细胞间隙增加、数目减少,细胞核固缩,染色强度加深。结论妊娠期甲减补充甲状腺激素治疗,增加了子代的体重,提高了学习记忆能力,增加了海马Reelin蛋白表达。对减少子代智力低下有重要意义。Objective:To explore the effect of thyroid hormone on the expression of rat behavior and Reelin protein in pregnancy. Methods:The female rats were randomly divided into normal group,thyroid hormone group and model group of 16,and the normal group was fed to the offspring(PN0). The other two groups during GD6-GD14 propylthiouracil,thyroid hormone group added GD15 to seed rat was born in thyroid hormone(L-T4),model group of ordinary feed to lactation was born,40 young mice in each group. GD6 and GD15,PN0 and PN7 weight of pregnant rats were measured,and serum thyroid hormone levels were detected in pregnant mice,and the test was conducted to locate the Reelin protein content. Results:In normal group,the TT4 was significantly higher than the TSH,which was significantly lower than the thyroid hormone group and the model group. The difference was statistically significant P0.05,indicating the success of the model. The weight of pregnant rats in the GD15 thyroid hormone group and the model group were significantly lower than that in the normal group. The PN0 and PN7 model group were significantly lower than normal group and thyroid hormone group,and the difference was statistically significant P0.05. On the first day,the second day,and the third day,the model group was significantly longer than the normal group and thyroid hormone group during the incubation period of the model group,and the difference was statistically significant P0.05. Reelin protein PN7,normal group(64.23 + 7.58)compared with thyroid hormone group(62.38 + 7.62),there was no obvious difference statistically significant(P0.05,the model group(28.46 + 9.33)significantly lower than the normal group and thyroid hormone group,difference was statistically significant(P0.05. The cell layers were disorganized,sparse and irregular,and the cell gap increased,the number decreased,the nucleus was fixed,and the dyeing intensity was deepened.
