摘 要:目的:探讨葫芦巴碱对HepG2细胞胰岛素抵抗的影响。方法:培养HepG2细胞,MTT法检测葫芦巴碱对HepG2细胞增殖的影响以确定可作用于HepG2细胞的葫芦巴碱的浓度;将HepG2细胞置于含胰岛素浓度为1×10^(-6)mol/L的培养液中,培养36 h成功建立胰岛素抵抗模型后,每孔加入100μL不同浓度的含药培养基,用葡萄糖检测试剂盒检测葫芦巴碱对胰岛素抵抗HepG2细胞葡萄糖消耗的影响。结果:葫芦巴碱浓度为3,000、2,500、2,000、1,500、1,000、500μmol/L时的细胞存活率分别为21.38%、39.23%、51.43%、73.36%、86.23%、91.88%,葫芦巴碱浓度>1,000μmol/L时对细胞有明显的毒性,故选择终浓度为1,000、500、250、100、50、25μmol/L进行实验,各浓度葫芦巴碱对胰岛素抵抗的HepG2细胞的葡萄糖消耗分别为(4.679±0.04)、(3.994±0.062)、(3.683±0.094)、(3.633±0.120)、(3.619±0.119)、(3.503±0.128)μmol/L,其中,前两个浓度与模型组比较,差异有统计学意义(P<0.05)。结论:葫芦巴碱对HepG2细胞胰岛素抵抗有改善作用。Objective: To investigate the effect of trigonelline on the insulin resistance of HepG2 cells. Methods: HepG2 cells were cultured in vitro. MTT assay was used to detect the effect of trigonelline on the proliferation of HepG2 cells. HepG2 cells were cuhured in medium with insulin concentration of 1×10-6 tool/L, and the insulin resistance model was established successfully for 36 h, then added 100 μL with different concen- tration of drug medium . The insulin resistance test was carried out by using the glucose detection kit to detect insulin resistance HepG2 Cell glucose consumption. Results: The Cell viability was 21.38%, 39.23%, 51.43%, 73.36%, 86.23% and 91.81% while the concentration of trigonelline was 3,000, 2,500, 2,000, 1,500, 1,000 and 500 μmol/L. There was obvious toxicity to ceils when the concentration is 〉 1000 mumol/L^-1, so the medium with concentration of 1000, 500, 250, 100, 50 and 25 μmol/L has been chosen. The glucose consumption on model HepG2 cells was (4.679±0.04), (3.994±0.062), (3.683±0.094), (3.633±0.120), (3.619±0.119) and (5.503±0.128)μmol/L. The difference between the first two concentrations is statistically significant compared with the model group. Conclusion: Trigonelline can improve insulin resistance in HepG2 cells.
