摘 要:【目的】研究中等强度持续训练对大鼠骨骼肌总抗氧化能力(total antioxidant capacity,T-AOC)、氧化产物丙二醛(malonaldehyde,MDA)、羟自由基(·OH)、过氧化氢酶(catalase,CAT)及谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性的影响,探讨对大鼠骨骼肌抗氧化功能的改善作用。【方法】采用SD大鼠跑台运动模型,进行中等强度持续运动训练,按照训练时间分为2、4、6周组,并设立正常对照组。各组大鼠训练结束后用匀浆法制备骨骼肌匀浆介质,分光光度法测定骨骼肌T-AOC、MDA、CAT、GSH-Px活性以及·OH抑制力。【结果】训练2周时,MDA和·OH抑制能力在水平高于对照组(P<0.05),训练6周组MDA含量较训练4周组显著降低(P<0.05),训练4周和6周后·OH抑制能力相比训练2周时下降(P<0.05)。训练4周和6周后CAT、GSH-Px水平高于对照组(P<0.05),6周时GSH-Px水平与4周相比差异无统计学意义。训练4周和6周后,T-AOC水平高于对照组(P<0.05),且训练6周组T-AOC水平高于训练4周组(P<0.05)。【结论】中等强度持续运动训练对大鼠骨骼肌抗氧化功能有较大影响,可以影响T-AOC、MDA、·OH抑制力、CAT及GSH-Px的水平,长期坚持中等强度持续运动训练有利于改善骨骼肌抗氧化损伤的能力。【Objective】To study the effects of moderate-intensity continuous exercise on the activities of total antioxidant performance(T-AOC),malondialdehyde(MDA),hydroxyl radical(·OH),catalase(CAT) and glutathione peroxidase(GSH-Px) in rat skeletal muscle,and explore the improvement of antioxidant function in rats.【Methods】SD rats were used to establish moderate-intensity continuous exercise training models with treadmill exercise methods.They were divided 4 groups:control group,2 w group,4 w group and 6 w group,according to the training time.Homogenate medium of skeletal muscle in each group was prepared by the homogenate method at the end of training.The activities of T-AOC,MDA,CAT,GSH-Px and the inhibition of·OH in skeletal muscle were measured by spectrophotometry.【Results】The level of MDA and the inhibition ability of·OH in the 2 w group were higher than those in the control group(P 0.05).The level of MDA in the 6 w group was significantly lower than that in the 4 w group(P〈0.05).The inhibition ability of·OH in the 4 w and 6 w groups was lower than that in the 2 w group(P〈0.05).The levels of CAT and GSH-Px in the 4 w and 6 w groups were higher than that in the control group(P〈0.05).The level of GSH-Px showed no significant difference between the 4 w and 6 w groups.The levels of T-AOC in the 4 w and 6 w groups were higher than that in the control group(P〈0.05).Moreover,the level of T-AOC in the 6 w group was higher than that in the 4 w group(P〈0.05).【Conclusion】Moderate-intensity continuous exercise has great influence on the antioxidant ability of rat skeletal muscle and can affect the levels of T-AOC,MDA,CAT,GSHPx and the inhibition ability of·OH.Long-term persistence of moderateintensity continuous exercise is beneficial to promote the ability of skeletal muscle against oxidative injury.
