摘 要:目的探讨不同时间点动脉泵注尤瑞克林(HUK)对动脉闭塞大鼠脑损伤的影响。方法将60只大鼠随机分为6组,分别为:假手术组(Sham组,n=10)、永久性中动脉闭塞(MCAO)模型组(MCAO组,n=10)、MCAO+生理盐水注射组(Na Cl组,n=10)、建模后0.5 h泵入HUK组(0.5 h组,n=10)、建模后1.5 h泵入HUK组(1.5 h组,n=10)和建模后3.0 h泵入HUK组(3.0 h组,n=10)。采用神经功能缺损评分、梗死面积测定、酶联免疫吸附试验和水迷宫实验评价MCAO大鼠脑损伤情况。结果与Sham组比较,MCAO组和NaCl组神经功能评分明显上升,脑梗死程度严重,脑脊液缓激肽(BK)及血管内皮生长因子(VEGF)含量增高,记忆力降低,组间比较,差异有统计学意义(P<0.05)。与MCAO组比较,0.5 h组、1.5 h组、3.0 h组的神经功能评分下降,脑梗死面积减少,脑脊液BK和VEGF含量降低,记忆力恢复,并且,随给药时间的延长,保护作用明显降低,组间比较,差异有统计学意义(P<0.05)。结论 HUK对动脉闭塞诱导的大鼠脑损伤有保护作用,且给药时间越早,保护作用越明显。Objective To investigate the effect of intra-arterial infusion of human urinary kallidinogenase( HUK) at different time points on brain injury in rats with arterial occlusion. Methods A total of 6 rats were randomly divided into 6 groups: the sham operation group( Sham group,n = 10),the middle cerebral artery occlusion( MCAO) model group( MCAO group,n = 10),MCAO + saline group( Na Cl group,n = 10),the group that HUK were injected after modeling 0. 5 hour( 0. 5 hour group,n = 10),the group that HUK was injected after modeling 1. 5 hours( 1. 5 hours group,n = 10) and the group that HUK was injected after modeling 3. 0 hours( 3. 0 hours group,n = 10). Neurological deficit score,infarct size,ELISA and water maze were used to evaluate the brain injury in rats with arterial occlusion. Results Compared with the Sham group,the score of neurological function in MCAO group increased significantly,the cerebral infarction was severe,the content of bradykinin( BK) and vascular endothelial growth factor( VEGF) in cerebrospinal fluid increased,and the memory was reduced,and the differences between the two groups were statistically significant( P〈0. 05).Compared with MCAO group,after intra-arterial infusion of HUK,the scores of neurological function reduced,the area of cerebral infarction reduced,the content of BK and VEGF in cerebrospinal fluid reduced,the recovery of memory recovered,and the protective effect reduced significantly with the delay of administration in the groups of 0. 5 hour,1. 5 hours and 3. 0 hours,and the differences were statistically significant( P〈0. 05). Conclusion HUK has a protective effect on cerebral injury induced by arterial occlusion in rats,and the earlier the protective effect is more obvious.
