摘 要:为有效降低烟叶中的淀粉含量、提高烟叶品质,从不同产地原烟表面初步分离具有降解淀粉能力的菌株,通过测定菌株的淀粉酶活性筛选高效产淀粉酶菌株,并对筛选菌株基因组DNA进行16S rDNA测序,同时结合生理生化试验对其进行鉴定。结果表明:在分离得到的22株菌株中,XC-3菌株产淀粉酶活性最高(116 U/mL);根据其16S rDNA测序结果以及生理生化试验结果推断,XC-3为苏云金芽孢杆菌(Bacillus thuringiensis)。进一步采用正交试验优化XC-3菌株在烟叶中的发酵条件,并考察其对烟叶中淀粉的降解效果。结果显示,将培养后的XC-3菌液(OD600=2.0)按照3%的烟叶质量比例接种于抽梗后的烟叶表面,在37℃条件下发酵48 h后,烟叶中的淀粉降解率可达29.84%,烟叶的总糖、还原糖含量上升,有效提升了烟叶的整体质量。In order to improve the tobacco quality by degrading the starch,several starch-degrading bacteria were isolated from the surface of tobacco leaves. The high-efficiency starch-degrading strain was obtained according to the assay of starch activity,and was identified by sequencing of the 16 S rDNA and physiological and biochemical tests. The results showed that strain XC-3 owned the highest activity of amylase-producing( 116 U/mL) among all 22 isolated bacteria strains. Strain XC-3 was identified as Bacillus thuringiensis according to the 16 S rDNA sequence and physiological and biochemical tests. The orthogonal experiments suggested that adding 3% culture medium with strain XC-3( OD600= 2. 0) to the surface of tobacco leaves followed by 48 hours fermentation under 37 ℃,the starch degrading rate could reach 29. 84%,and the contents of total sugar and reducing sugar were both increased,hence the tobacco quality was improved.
