摘 要:目的探讨直流电刺激(t DCS)对卒中模型大鼠脑组织神经递质的影响。方法取大鼠随机分成3组,即对照组、模型组、实验组,每组15只。除对照组外,其他各组施以手术,制作大鼠MCAO模型,建立起卒中模型后1 w,每天给予电刺激组经颅直流电刺激。造模4 w断头,取卒中侧大脑,快速冰冻,冰箱保存。观察各组大鼠神经组织病理学变化,并以高效液相色谱法测定各组大鼠脑组织中单胺类神经递质5-羟色胺(5-HT)和多巴胺(DA)、5-羟吲哚乙酸(5-HIAA)、3,4-二羟基苯乙酸(DOPAC)的含量及其对大鼠运动功能的影响。结果与模型组比较,DA、DOPAC含量均显著升高,差异有统计学意义(P<0.05或P<0.01),而5-HT、5-HIAA变化不明显;并且观测到大鼠平衡木测验和网屏测验结果提示电刺激组功能优于模型组。结论通过t DCS可使卒中模型大鼠脑组织中单胺类神经递质的含量提高而影响脑的神经传导和大脑神经功能的修复与重建。Objective To study the effects of transcranial direct current stimulation on neurotransmitter in brain tissues of rats with cerebral apoplexy. Methods Rats were randomly assigned to control group,model group,experimental group with each group of 15 rats. In addition to control group,other groups were operated in order to establish MCAO( middle cerebral artery occlusion) model. After 1 week of modeling,transcranial direct current stimulation was given a day. Rats were beheaded after 4 weeks of modeling. Lateral brain was removed and then frozen quickly in the refrigerator. Pathology change in rats' nerve tissue was observed,with high performance liquid chromatography( HPLC) method in determining content of monoamine neurotransmitter serotonin( 5-HT) and dopamine( DA),5-hydroxyl insole acetic acid( 5 HIAA),3,4-2hydroxyl benzene acetic acid( DOPAC) and its impact on motor function of rats. Results Compared with model group,the content of DA and DOPAC were significantly increased( P 0.05 or P 0.01),however,5-HT and 5-HIAA change was not obvious,and the results of the balance beam test and screen test indicated electrical stimulation group function was better than model group's. Conclusion The level of monoamine neurotransmitters in brain tissue of rats with cerebral apoplexy is increased by t DCS,which affect the nerve conduction and repair and reconstruction of brain function.
