摘  要:[目的]探讨白藜芦醇治疗结肠癌的作用机制。[方法]以10mg/ml浓度的白藜芦醇处理HCT116细胞,进行损伤愈合实验及克隆集落实验,检测白藜芦醇对结肠癌细胞株迁移及增殖能力的影响。使用Cytation5细胞成像微孔板检测仪动态检测HCT116细胞增殖情况。通过流式细胞术检测白藜芦醇对HCT116细胞凋亡以及对细胞周期的影响。运用Western Blot技术检测白藜芦醇处理后NF-κB蛋白的表达情况。[结果]给予白藜芦醇24h后HCT116细胞迁移能力减弱,增殖减少,凋亡增加,并且使细胞周期阻滞于G2期,蛋白NF-κB表达减少。[结论]白藜芦醇抑制HCT116细胞的迁移和增殖,并且促进凋亡及阻滞其周期于G2期,同时NF-κB蛋白表达降低,提示白藜芦醇上述抗癌效应与NF-κB通路密切相关。[Objective]To provide significant insights into the molecular mechanism of resveratrol against colon cancer.[Methods]HCT116 cells were treated at a concentration of 10 mg/ml resveratrol.Performing wound healing assay and clonogenic assay,we detected the migratory and proliferative properties of resveratrol on HCT116 cells.Using Cytation 5 Cell Imaging Multi-Mode Reader,the proliferation of cells was dynamically examined.The apoptosis and cell cycle of HCT116 cells treated with resveratrol were studied by flow cytometry.The relative expression of NF-κB was conducted by Western Blot.[Results]The ability of migration of HCT116 cells treated with resveratrol weakened,proliferation reduced,apoptosis increased,cell cycle arrested in G2 phase,and the expression of NF-κB reduced.[Conclusion]Resveratrol-mediated apoptosis and inhibition of cellular growth and proliferation of HCT116 cells were associated with NF-κB signaling pathway.
