摘  要:目的评价根管充填材料在不同浓度及时间下的细胞毒性。方法将ParaCore树脂、ParaBond粘接剂、牙胶、AH Plus根管封闭剂分别放置1 d(新鲜组)和7 d(陈旧组),制取浸提液,并制成100.0%、50.0%、25.0%、12.5%的稀释液,与L-929细胞接触24 h,采用二甲基噻唑二苯基四唑嗅盐比色法,比较不同材料在不同浓度和时间下的相对增值率(RGR)。结果新鲜组4种浓度的稀释液中,ParaCore树脂的RGR大于牙胶,ParaBond的RGR小于AH Plus,差异有统计学意义(P均<0.05)。陈旧组4种浓度的稀释液中,ParaCore的RGR大于牙胶,除12.5%浓度外,差异有统计学意义(P均<0.05);ParaBond的RGR小于AH Plus,除25.0%、12.5%浓度外,差异有统计学意义(P均<0.05)。在新鲜组中,随着浸提液浓度增大,各材料RGR降低,不同浓度下,ParaCore、ParaBond和AH Plus的RGR两两比较,差异有统计学意义(P均<0.05)。放置时间越长,RGR越大,除牙胶外,不同时间下其他3种材料RGR差异有统计学意义(P均<0.05)。结论 ParaCore树脂的细胞毒性小于牙胶,AH Plus封闭剂的细胞毒性小于ParaBond粘接剂;4种材料的毒性具有浓度相关性;除了牙胶,其他3种材料具有时间相关性。Objective To evaluate the cytotoxicity of root filling materials at different concentrations and time. Methods Four kinds of materials (ParaCore bonding resin, ParaBond adhesive, gutta-percha, AH -Plus endodontics sealer)were set for 1 day (fresh samples) and 7 days (aged sam- ples). Four solutions of 100. 0% , 50. 0% , 25.0% , 12. 5% material concentrations were diluted respectively for every material and then were incubated with the L - 929 fibroblasts for 24 hours. The cytotoxicity of the materials was assessed by cell viability using the bromide 3 - (4,5 dimethylthiazol -2yl) -2, 5 -diphenyhetrazolium assay method. Results In four concentrations of fresh samples, the cell viability of ParaCore was significantly higher than gutta - percha ( P 〈 0. 05 ), and the cell viability of ParaBond was significantly lower than AH Plus(P 〈 0. 05). In four concentrations of aged samples, except the 12. 5% concentra- tion group, ParaCore was significantly higher than gutta - percha in the other three groups ( P 〈 0. 05 ). The cell viability of ParaBond was significantly lower than AH Plus in the 100. 0% and 50. 0% concentration groups(P 〈0. 05 ). In fresh samples, the greater the concentratio,the lower the cell viability. The differences between every two groups of other three materials except gutta - percha were statistically significant(P 〈 0. 05 ). With the increase of setting time, the cell viability increased in all materials. There was statistical significance between fresh and aged groups of all materials except gutta - percha ( P 〈 0.05 ). Conclusion The ParaCore has a lower cytotoxicity than gutta - percha and the ParaBond has higher cytotoxicity than AH Plus. The concentration had an influence on the cytotoxicity of all tested materials. Setting time had an influence on the cytotoxicity of other tested materials except gutta-percha.
