摘  要:目的:评价国内西伯利亚蓼资源的遗传分化程度,为种群分化、物种进化、资源保护和药材品质保障提供科学依据。方法:从国内4个省份13个采样点采集56份西伯利亚蓼,开发并筛选多态性EST-SSR标记,结合相关序列扩增多态性(SRAP)分子标记扩增结果,通过计算遗传距离和聚类分析,评价不同生态分布区域内西伯利亚蓼的遗传分化程度。结果:开发的45对EST-SSR引物中,38对引物有扩增产物,共扩增出660个条带,其中17对引物产生的多态性条带比率为100%,共扩增出多态性条带303条。96对SRAP通用引物中,67对引物有扩增产物,共扩增出1 347个条带,其中58对引物产生的多态性条带比率为100%,共扩增出多态性条带1 147条。种群遗传距离计算和聚类分析结果表明,西伯利亚蓼种群内和种群间均有遗传分化,分布区域相隔越远,遗传分化程度越高。结论:地理位置隔离、海拔高度均影响了国内西伯利亚蓼种质资源的分布和分化,人类活动可能也对其有影响。Objective: To explore the genetic differentiationof Polygonum sibiricum in different areas in China,in order toprovide basis for its population subdivision,evolution,protection and utilization,and quality control. Method: Fiftysix samples were collected from 13 sampling sites in four provinces of China. EST-SSR primers were developed and screened,and the genetic differentiation was evaluated with both EST-SSR and sequence related amplified polymorphism( SRAP) markers. Result: Among 45 pairs of EST-SSR primers,38 pairs amplified 660 distinguished bands,and 17 pairs amplified 303 polymorphic bands,with a polymorphism rate of 100%. Among 96 pairs of SRAP primers,67 pairs amplified 1 347 distinguished bands,and 58 pairs amplified1 147 polymorphic bands,with a polymorphism rate of 100%. Genetic distance and cluster analysis revealed both inter-and inner-population have genetic differentiation. Conclusion: Genetic variation and geographic differentiation areobserved among P. sibiricum populations in China,which were affected by geographic distances,altitudes and possibly mankind activities.
