摘  要:目的研究维甲酸相关孤儿受体(RORα)与孤儿核受体(REV-ERBα)在人正常胃组织和胃癌组织中的表达情况、相关性及其临床病理意义。方法收集50例不同临床病理分期的胃癌患者的石蜡切片,运用免疫组织化学染色技术检测RORα与REV-ERBα在人正常胃组织及不同临床病理分期胃癌组织中的表达,同时Western blot研究RORα与REV-ERBα在人正常胃组织及不同TMN分期人胃癌组织中的表达情况。结果免疫组织化学染色结果显示,RORα与REV-ERBα在人胃癌组织中表达明显降低,同时RORα与REV-ERBα的表达水平与胃癌患者TMN分期(P=0.013、0.010)及有无淋巴结转移(P=0.001、0.002)密切相关,与患者年龄、性别及肿瘤大小无相关性。Mc Nemar检验显示RORα与REV-ERBα在人胃癌组织中的表达呈正相关性(Kappa=0.377,P=0.007)。同时Western blot结果显示RORα与REV-ERBα在人胃癌组织中表达明显下降,且与TMN分期有显著相关性(P<0.05)。结论 RORα与REVERBα在人胃癌组织中表达降低与人胃癌组织的TMN分期及有无淋巴结转移有密切关联,并且RORα与REV-ERBα可能在人胃癌组织中存在协同表达及调控,从而影响胃癌的发生与发展。Objective To research the retinoid - related orphan receptor alpha (RORct) and nuclear receptor nuclear clinical paraffin receptor subfamily 1 ,group D member 1 (NrlD1 ,also known as REV-ERBα) expression,corelation and pathological significance in human normal gastric tissues and gastric cancer tissues. Methods Collected 50 specimens with human normal gastric tissues and gastric cancer tissues. The expression of RORα and REV- ERBα in human normal gastric tissues and different clinical pathological stages were detected by immunohistochem- ical staining and Western blot. Results The expression of RORα and REV-ERBα in human gastric cancer tissues were reduced. They were significant in gastric cancer TMN stage (P = 0. 013,0. 010) and lymph node metastasis (P =0. 001,0. 002) ,however,age,sex and tumor size had no statistical difference. Additional,there was a positive correlation between RORα and REV-ERBα in human gastric cancer tissues by McNemar analysis ( Kappa = 0. 377, P = 0. 007 ). Western blot results demonstrated that the RORα er tissues with increased TMN stage (P 〈 0.05 ). Conclusion and REV-ERBα was reduced in human gastric canc- RORα and REV-ERBα are closely associated with TMN staging and lymph node metastasis in human gastric cancer tissues, and RORα and REV-ERBα may be syner- gistically expressed and regulated in human gastric cancer tissues, which may affect the expression of RORα and REV-ERBα in human gastric cancer occurrence and development.
