摘  要:目的 :探讨高强度聚焦超声 (HIFU)固化疫苗对荷 H2 2 肝癌小鼠免疫功能的影响。方法 :15 0只昆明小鼠随机分为三组 :生理盐水组 (A组 ) ,高温固化 H2 2 瘤苗组 (B组 ) ,HIFU固化 H2 2 疫苗组 (C组 )。于第 1、 2、 3、4周 ,各组小鼠分别注射生理盐水 ,高温固化瘤苗 ,HIFU固化疫苗。第 5周 ,各小鼠皮下接种同源 H2 2 癌细胞 1× 10 6个 ,记录各组小鼠肿瘤发生率、肿瘤体积、T淋巴细胞亚群及 CD6 8细胞数。结果 :A、B、C三组肿瘤发生率分别为10 0 % ,80 % ,4 5 % ,C组较 A、B两组显著降低 (P<0 .0 5 ) ,C组肿瘤体积较 A、B两组显著减小 (P<0 .0 1) ,C组CD4 + / CD8+比值高于生理盐水组 (P<0 .0 1) ,CD6 8+与其他两组相比有显著性差异 (P<0 .0 1)。结论 :HIFU固化疫苗有较强的免疫效应 ,是一种良好的肿瘤疫苗。Objective:To evaluate the immune effect of high intensity focused ultrasound(HIFU)coagulation tumor vaccine on mouse transplanted with H 22 hepatic carcinoma.Methods:150 KM mouse were divided into three groups:normal saline group(group A),thermal coagulation H 22 carcinoma vaccine(group B),and HIFU coagulation H 22 carcinoma vaccine(group C).The three groups were inoculate normal saline,thermal coagulation H 22 carcinoma vaccine,HIFU coagulation H 22 carcinoma vaccine respectively in the I st ,2 nd ,3 rd ,4 th week.In the 5 th week,the three groups were inoculated subcutaneously with 1×10 6 H 22 hepatic carcinoma cells.The cancer incidence and tumor volume of inoculated tumor were observed,the T lymphocyte subset and the number of CD68 + cell in every mouse were tested.Results:The cancer incidence in group A,B,C were 100%,80%,45% respectively,which was significantly lower in group C( P <0 05).The volume of tumor in group C was significantly smaller than that in group A and B( P <0 001).The rate of CD 4 +/CD 8 + in group C is higher than that in group A( P <0 01),also the number of CD 68 + cell in group C was more than that in group B and A.Conclusions:HIFU coagulation tumor vaccine was more effective than thermal coagulation tumor vaccine.
