摘  要:目的:通过蛋白质组学研究,检测面神经钳夹损伤后的大鼠面神经与正常大鼠面神经的差异蛋白。方法:分别取正常大鼠面神经和钳夹损伤30秒后的大鼠面神经,分别作为对照组和实验组。提取两组大鼠面神经蛋白后,质谱分析并鉴定筛选出2组大鼠面神经组织差异表达的蛋白。采用SPSS15.0软件包对数据进行统计学分析。结果:经蛋白质组学研究,成功鉴定出19个2组间显著性差异蛋白,其中4个蛋白点差异倍数在20倍以上。4个蛋白主要涉及代谢、信息储存以及信号转导3大功能。结论:面神经钳夹损伤后与正常大鼠面神经组织间存在差异蛋白,有助于探讨面神经损伤后的蛋白调控机制。Objective: To investigate the differentially expressed proteins of facial nerve between normal rats and facial nerve crush injury rats. Methods: Facial nerve and crush injury nerve were selected as normal controls control group and experimental group respectively. After protein extraction, differentially expressed proteins were separated by two-dimensional gel electrophoresis (2-DE) and identified by MALDI-TOF/TOF. Statistical analysis of data was performed using SPSS 15.0 software package. Results: According to the proteomics study, there were 19 significant differences between the two groups, and the difference of 4 protein points was more than 20 times. The 4 proteins fell into 3 functional categories as follows: metabolism, information storage and transduction. Conclusion: The differentially expressed proteins of facial nerve between normal rats and facial nerve injury rats may be new targets for investigating the mechanisms of the effect of nerve injury.
