摘 要:Kisspeptin是调控脊椎动物生殖的重要神经内分泌因子,本实验室先前已鉴定了褐牙鲆kiss2及其受体gpr54-2基因,二者在褐牙鲆脑和性腺中具有较高表达,但其在褐牙鲆早期性腺分化中的作用以及温度、外源激素对其基因表达的影响尚不清楚。因此本研究设置高温(28℃)、雌激素(10μg/L)、雄激素(10μg/L)处理组和常温(20℃)对照组培育性腺分化早期的褐牙鲆幼鱼(45日龄、55日龄和65日龄),利用荧光定量PCR技术分析了高温和性激素诱导对褐牙鲆早期性腺分化过程中kiss2及其受体gpr54-2基因表达的影响。试验结果显示,高温和雌激素处理可以促进褐牙鲆脑中kiss2和性腺中gpr54-2的表达,却下调了褐牙鲆性腺中kiss2的表达;而雄激素处理则使褐牙鲆脑和性腺中kiss2和gpr54-2的表达几乎都受到显著的抑制作用。本研究结果为进一步阐述kiss2和gpr54-2基因在褐牙鲆早期性腺分化中的作用奠定基础。Kisspeptin is an important neuroendocrine factor for the regulation of vertebrate reproduction.The kiss2 and its receptor(gpr54-2)cDNAs have been identified previously and are highly expressed in the brain and gonads of olive flounder Paralichthys olivaceus.However,the function of kiss2 and gpr54-2 in the early gonadal differentiation and the effects of temperature and exogenous hormones on the genes expression are unclear.Juvenile flounders(45 days post-hatch dph,55 dph and 65 dph)were cultured in four experimental groups of high temperature(28 ℃),estrogen(10μg/L),androgen(10μg/L)and normal temperature(20 ℃),and the effects of high temperature and sex hormone on the expression of kiss2 and its receptor gpr54-2 were analyzed by qPCR.The results showed that the high temperature and estrogen treatment led to promote the expression of kiss2 in brain and gpr54-2 in gonad of olive flounder,but downregulated the expression of kiss2 in gonad of the flounder,while the androgen treatment almost significantly inhibited the kiss2 and gpr54-2 expression.The findings provide a basis for further understanding of the role of kiss2 and gpr54-2 genes in the early gonadal differentiation in olive flounder.
