摘  要:为了研究西藏工布乌头(Aconitum kongboense L.)种质的遗传多样性,探讨西藏工布乌头的亲缘关系,试验采用叶绿体微卫星(cpSSR)分子标记技术对西藏工布乌头的遗传多样性进行分析。结果表明:通过筛选得到6对cpSSR引物,其中2对引物的扩增片段显示有突变位点。15份西藏工布乌头非编码区trnS-trnfM多态性最高,其单倍型多样性(Hd)、核苷酸多样性(Pi)分别为0.467,0.000 73。Tajima’s D检验中所有区域的D值均未达到显著水平(P〉0.05),自然选择对这些突变并不起作用。HAP3与HAP1、HAP2、HAP4的亲缘关系都比较远。说明西藏工布乌头种质的遗传多样性较低,加查县崔久乡和浪卡子县卡热乡两个地区的野生工布乌头显示出较近的亲缘关系。The aim of the present paper was to systematically investigate the genetic diversity of Tibetan Aconitum kongboense L..The genetic diversity of Aconitum kongboense L.in Tibet was analyzed by using chloroplast microsatellite(cpSSR) molecular marker technology.The results showed that six pairs of cpSSR primers were obtained by screening,and the amplified fragments of 2 pairs of primers showed mutation sites.The polymorphism of non-coding region trnS-trnfM was the highest in 15 samples of Tibetan Aconitum kongboense L..The haplotype diversity(Hd) and nucleotide diversity(Pi) of trnS-trnfM were 0.467 and 0.000 73,respectively.All the D values of Tajima's test were not significant(P〉0.05),indicating that natural selection had no effect on these mutations.Mediation network showed that The genetic relationships were far between HAP_3 and HAP1,HAP2,HAP4,respectively.Conclusively,the genetic diversity of Tibetan Aconitum kongboense L.was low,and the genetic relationship of the wild Aconitum kongboense L.from Cuijiu Township in Jiacha County and Kare Township in Longqiazi County showed relatively close.
