基质金属蛋白酶-13、基质金属蛋白酶-14和组织蛋白酶D在结肠癌中的表达及意义

摘　　要：目的探讨基质金属蛋白酶（metalloproteinase，MMP）-13、MMP-14及组织蛋白酶-D（Cathep—sinD）在结肠癌中的表达及意义。方法收集2014年2月至2016年3月内蒙古医科大学附属医院80例结肠癌患者的手术切除组织，对其进行染色判定，同期选择80例我院健康体检者正常黏膜标本作为对照组。观察MMP-13、MMP-14、CathepsinD在结肠癌临床病理特征的关系，两种结肠组织MMP-13、MMP-14、CathepsinD的表达情况。结果结肠组织中MMP-13、MMP-14、CathepsinD性别、年龄、肿瘤部位无统计学意义（P〉0．05），肿瘤大小、组织学分级、Dukes分期、淋巴转移有统计学意义（P〈0．05）；结肠癌组织中的MMP-13、MMP-14、CathepsinD阳性表达率显著高于对照组[75．00％（60／80）比1．25％（1／80）、87．50％（70／80）比12．50％（10／80）、71．25％（57／80）比15．00％（12／80）（P〈0．05）]。结论MMP-13、MMP-14、CathepsinD的阳性表达在结肠癌组织与正常组织中存在差异，MMP-14与结肠癌的分化程度、分期、浸润转移之间密切相关，联合检测MMP-13、MMP-14、CathepsinD的表达可能是判断病情和预后的重要指标。Objective To investigate the expression and significance of matrix metalloproteinase-13 （MMP-13） ,matrix metalloproteinase-14（MMP-14） and Cathepsin D in colon cancer. Methods Eithty surgical resection tissues of colon cancer patients were collected for immunohistochemical staining in Affiliated Hospital of Inner Mongolia Medical University from February 2014 to March 2016. At the same time,80 mucosal tissues of healthy controls were also collected. The expression of MMP-13 ,MMP-14 and Cathepsin D both in colon tis- sues and healthy tissues were all analyzed. Results The expression of MMP-13, MMP-14 and Cathepsin D had no correlation with the sex, age, and tumor site （ P 〉 0.05 ）. While they were closely related with the tumor size, histological grade, Dukes stage, and lymph node metastasis（ P 〈 0.05 ）. The positive expression rate of MMP-13, MMP-14 and Cathepsin D in colon cancer tissues were significantly higher than in healthy control tissues [75.00% （60/80） Vs. 1.25% （ 1/80）, 87. 50% （70/80） Vs. 12. 50% （ 10/80）, 71. 25% （57/80） Vs. 15.00% （12/80） ] （ P 〈 0.05 ）. Conclusion There are significant differences between colon cancer tissues MMP-13 ,MMP-14 and Cathepsin D positive expression rate in normal tissue ,the degree of differentiation,MMP- 14 and colon cancer staging, metastasis is closely related between the expression of combined detection of MMP- 13, MMP-14 and Cathepsin D may be an important index to judge the severity and prognosis.
