摘 要:目的探讨肝激酶B1(LKB1)在胸腺癌及癌旁组织中的表达及其在胸腺癌细胞生长与转移中的作用及分子机制。方法选取海军军医大学附属长海医院自2009年1月至2017年12月收治的58例侵袭性胸腺瘤及胸腺癌患者的胸腺癌组织及癌旁组织样本为研究对象。利用免疫组织化学方法检验癌组织与癌旁组织中LKB1的表达水平;构建稳定过表达LKB1的人胸腺癌MP59和T1889细胞系,利用平板克隆实验检测LKB1对胸腺癌细胞增殖的影响;利用Transwell实验检测LKB1对胸腺癌细胞迁移侵袭的影响;通过蛋白质印记(Western blot)筛选LKB1调控胸腺癌细胞生长及转移的下游信号通路。结果 81.0%(47/58)胸腺癌组织中LKB1的表达水平低于癌旁组织(P<0.05)。LKB1过表达抑制胸腺癌细胞MP59与T1889的生长和迁移能力,且上皮细胞钙粘蛋白(E-cadherin)分子被上调。结论 LKB1在胸腺癌中低表达,对胸腺癌的生长与转移具有抑制作用。Objective To investigate the expression of LKB1 in thymic carcinoma tissue and explore the function of LKB1 in thymus carcinoma and the underling molecular mechanism. Methods A sample of 58 patients with invasive thymoma and thymie carcinoma who were admitted to the hospital from January 2009 to December 2017 were selected as the study subjects. The expression of LKB1 in thymie carcinoma tissue was checked by using immuuohistochcmieal detection. Thymic carcinoma cell lines MP57 and T1889 were transfected with overexpression LKB1 plasmid. The proliferation ability of MP57 LKB1 or T1889 LKB1 and their control cells were checked by cloning formation assay. The metastasis ability of MP57 LKB1 or T1889 LKB1 and their control cells were checked byTran- swell experiment. To explore the downstream signaling pathways of LKB1 in thymie cancer cells proliferation and metastasis by western blot. Results The expression level of LKB1 in 81.0% ( 47/58 ) thymus carcinoma was lower than that of the adjacent tissues ( P 〈 0. 05 ). LKB1 overexpression inhibited the growth and migration of thymus cancer cells MP59 and T1889, and the E-cadherin were up- regulated. Conclusion LKB1 expressing is decreasing in thymie carcinoma. It could inhibit growth and metastasis of thymic carcino- ma cells via upregulating E-eadherin patways.
