摘 要:结直肠癌(colorectal cancer,CRC)是消化道最常见的恶性肿瘤之一,化疗是不可手术根治CRC患者的一线治疗方案,但细胞耐药的出现严重影响了其治疗和预后。自噬与肿瘤的发生、发展密切相关,介导肿瘤细胞耐药,近期研究结果表明,自噬与P-g P介导的肿瘤耐药相关,miRNA、肿瘤干细胞、c-Jun N-末端激酶、STAT3均可通过调控自噬增强CRC细胞的药物敏感性。因此,调控自噬有望成为逆转CRC细胞耐药的新靶点,抑制CRC细胞自噬联合化疗药物的应用,有望成为耐药CRC治疗的新策略,进一步深入研究,可为耐药CRC患者的临床治疗提供新思路。Colorectal cancer(CRC) is one of the most common malignant tumors of the digestive tract. Chemotherapy is the first-line treatment for patients without radical surgery,but the emergence of resistance to chemotherapy seriously affects the treatment and prognosis of patients. Autophagy is closely related to the occurrence and development of tumor and mediate resistance of tumor cell. Recent studies have shown that it is associated with P-gP-mediated tumor resistance. miRNA,cancer stem cells,c-Jun N-terminal kinase,STAT3 can regulate autophagy and enhance the drug sensitivity. Therefore,regulation of autophagy is expected to reverse the new target of resistance of CRC,inhibition of CRC cell autophagy combined with chemotherapy drugs,which is expected to become a new strategy for drug-resistant CRC.Further studies should be performed to provide sufficient evidences and new ideas for drug-resistant CRC treatment.
