摘 要:目的应用长链非编码RNA基因芯片研究HOX基因在喉癌组织与癌旁正常组织中的表达谱。方法设立5组经过严格配对的实验组(人喉癌组织)与对照组(同一患者癌旁正常组织)。手术中切取人喉癌组织及癌旁正常组织,应用Triozl法抽提所取标本总RNA,纯化RNA后,将等量的两组RNA样本逆转录为荧光标记的cRNA混合物,与长链非编码RNA基因芯片进行杂交。应用Agilent扫描仪对芯片荧光信号图像进行扫描,及对原始数据进行计算机统计学分析,结果得到实验组与对照组两种组织之间HOX基因的LncRNA和mRNA的差异表达谱。结果在LncRNA表达谱的统计结果中,筛选出差异表达HOX基因簇的LncRNA有18条,表达上调的有8条,表达下调的有10条。在mRNA表达谱的统计结果中,筛选出差异表达的HOX基因簇的mRNA有17条,表达上调的有15条,表达下调的有2条。结论与癌旁正常组织相比,HOX基因在喉癌组织中的表达谱发生了显著变化。Objective To study the expression profile of HOX gene in laryngeal carcinoma tissue and adjacent normal tissue by long-chain non-coding RNA gene chips. Method Five groups were divided into an experimental group (human laryngeal carcinoma tissue) and a control group (adjacent normal tissue of the same patients) by strict matching. Human laryngeal carcinoma tissue and adjacent normal tissue were cut out during surgery, and the total RNA was extracted by Triozl method. After RNA was purified, the RNA samples of the two groups were reverse transcribed into a mixture of fluorescently labeled cRNA and were hybridized with long-chain non-coding RNA gene chips. The fluorescence signal images of the chips were scanned by Agilent scanner, and the original data were statistically analyzed by computer. The differential expression profile of LncRNA and mRNA of HOX genesof the experimental group and the control group were obtained. Results In the statistical results of LncRNA expression profile, there were 18 LncRNAs screened with different expression of HOX gene cluster, with 8 of them up-regulated and 10 of them down-regulated. In the statistical results of mRNA expression profile, there were 17 mRNAs screened with different expression of HOX gene cluster, with 15 of them up-regulated and 2 of them down-regulated. Conclusion The expression profile of HOX gene in laryngeal carcinoma tissue was significantly changed compared with normal tissue.
