摘 要:采用高效液相色谱法测定诺丽果实中草酸、酒石酸、苹果酸、抗坏血酸和柠檬酸的含量。样品用磷酸二氢钾+磷酸水溶液经超声波提取,用SinoChrom ODS-BP色谱柱分离,流动相为磷酸二氢钾+磷酸溶液pH2.8,流速0.6 m L/mi n。在该条件下中草酸、酒石酸、苹果酸、抗坏血酸和柠檬酸得到很好的分离,分别在1.0-2.5、1.0-2.5、1.5-2.5和1.0-2.5μg的范围内有良好的线性范围,相关系数R^2分别为0.999 0、1.000 0、0.999 9、0.999 9和1.000 0,最低检出限分别为1.0、2.0、2.5、1.5和3 mg/kg,加标回收率分别为101.3%、98.7%、97.3%、98.3%和97.5%。该方法快速、准确,能用于测定诺丽果实中有机酸的含量。Organic acid in Noni sample were determined by HPLC. Samples was extracted by phos- phate buffer solution and ultrasonic, separated on the SinoChrom ODS-BP column using phosphate buffer solution with pH 2.8 as the mobile phase, and the flow rate of 0.6ml/min. In these condition, oxalic acid, tartaric acid, malic acid, ascorbic acid and citric acid could be well separated. Organic acid showed a good linearity in 1.0-2.5, 1.0-2.5, 1.5-2.5 and 1.0-2.5 μg(R2=0.999 0, 1.000 0, 0.999 9, 0.9999, 1.0000), the LOD were 1.0, 2.0, 2.5, 1.5 and 3 mg/kg and the recoveries were 101.3%, 98.7%, 97.3 %, 98.3 % and 97.5 %. The verified results demonstrated that this method was precise and accu- rate and could be used for determination of organic acids in noni.
