摘 要:目的:观察电针对慢性应激抑郁模型大鼠海马和额叶cGMP、MDA和SOD的影响,进一步研究电针干预治疗抑郁症的生物学机制。方法:将32只健康雄性SD大鼠随机分对照组、模型组、电针组、氟西汀组,每组8只。除对照组外,其余各组大鼠进行孤养,并接受慢性不可预知性应激刺激方式造模。各组大鼠分别于应激前1 d、应激2 d后进行开野试验、测量体质量和糖水消耗试验,观察大鼠的行为学变化。应激21 d后,断头取脑,采用放射免疫法分别检测大鼠海马和额叶cGMP含量;黄嘌呤氧化酶法测海马、额叶SOD活性;硫代巴比妥酸法测海马、额叶MDA含量。结果:模型组大鼠行为学指标低于对照组(P<0.01);氟西汀组和电针组均高于模型组,大鼠行为学指标高于模型组(P<0.05)。与对照组比较,模型组大鼠海马和额叶组织cGMP、MDA升高(P<0.01),SOD降低(P<0.01)。与模型组比较,氟西汀组、电针组海马和额叶组织的cGMP、MDA含量均低于模型组(P<0.05),SOD的含量均高于模型组(P<0.05)。结论:电针可改善慢性应激抑郁大鼠的行为学变化,具有较好的抗抑郁作用;电针可降低模型大鼠不同脑区cGMP、MDA含量、升高SOD含量,这可能是电针治疗抑郁症的可能机制。Objective: To observe the effects of electro-acupuncture on cGMP, MDA and SOD in hippocam- pus and frontal lobe of chronic stress depression in rats, and to further study the biological mechanism of elec- tro-acupuncture intervention in the treatment of depression. Methods: Thirty-two healthy male Sprague-Dawley rats were randomly divided into 4 groups (n=8), respectively are normal group, model group, electro-acupuncture group and fluoxetine group. Except the normal control group, the rest of the rats were subjected to loneliness and subjected to chronie unpredictable stress stimulation. Rats in each group were subjected to open field test 1 day before stress and 21 days after stress. Body weight and sugar consumption test were measured to observe the behavioral changes of rats. After 21 days of stress, the brain was decapitated and the cGMP content of hip- poeampus and frontal lobe were detected by radioimmunoassay. The activity of SOD in hippocampus and frontal lobe was measured by xanthine oxidase method. The content of MDA in hippocampus and frontal lobe was measured by thiobarbituric acid method. Results: After 21 days, the behavioral indexes of the model group were significantly lower than those of the normal control group (P〈0.01). The behavior index of the rats in the fluox- etine group and the electro-acupuncture group was significantly higher than that in the model group (P〈0.05).Compared with the normal control group, the levels of cGMP and MDA in the hippocampus and frontal lobe of the model group were significantly increased (P〈0.01), and the SOD was significantly decreased (P〈0.01). Compared with the model group, the levels of cGMP and MDA in hippocampus and frontal lobe tissue were decreased (P〈0.05), and the content of SOD in fluoxetine group and electro-acupuncture group were lower than the model group (P〈0.05).
