摘  要:白色花被周边具不规则紫红色条纹的洒锦系品种是荷花种质资源的特殊类型,极具观赏价值。通过形态学和分子生物学手段揭示洒锦品种之间的亲缘关系和分子水平差异,有助于更好地开发利用这类资源。试验以传统的‘单洒锦’(Nelumbo nucifera‘Dan Sajin’)、‘大洒锦’(N.nucifera‘Da Sajin’)和‘小洒锦’(N.nucifera‘Xiao Sajin’)种质为材料,在调查其花、叶及结实性等10个表型性状基础上,比较217对高多态性EST-SSR引物扩增所得3者之间的差异条带及其分子生物学信息。结果显示:形态学上,与‘大洒锦’和‘小洒锦’相比,‘单洒锦’的花被片数少、花冠大、雄蕊多、立叶高大、结实率高;在分子水平上,‘大洒锦’和‘小洒锦’之间未获得差异条带,二者和‘单洒锦’共筛选出28对多态性引物,其差异条带比对得到相似度高、功能部分明确的基因20个,尚未被验证功能的高相似度基因4个。本研究表明:就群体性状而言,重瓣的‘大洒锦’和‘小洒锦’之间形态差异较小,两者与单瓣的‘单洒锦’区别较大;3个洒锦品种之间‘大洒锦’和‘小洒锦’亲缘关系较近,两者与‘单洒锦’遗传距离较远。Variegated-petalled cultivars in sacred lotus (Nelumbo nucifera) are well worth appreciating due to white perianth with irregular purple stripes around. It will help us to exploit and utilize the variegated resource by understanding the genetic relationship and molecular level differences between the cultivars by applying morphological and molecular biological methods. Three variegated cultivars of lotus, ‘Dan Sajin', ‘Da Sajin', ‘Xiao Sajin', were subjected to investigate the diversity on the phenotypic characters including 10 indexes such as flower, leaf and fruit-setting, and the difference of polymorphic bands and their biology information amplified by 217 pairs of EST-SSR primers among them. The results showed that ‘Dan Sajin' exhibited fewer perianths, bigger flower diameter, larger number of stamens, higher and bigger aerial leaves and higher fruit-setting than those of ‘Da Sajin' and ‘Xiao Sajin'. On the molecular level, there were no different bands between 'Da Sajin' and 'Xiao Sajin'. 20 genes with high ident and partially defined function, and 4 genes with high ident but unauthenticated function were blasted based on the polymorphic bands from the 28 pairs of EST-SSR primers among the three materials. In conclusion, our findings suggested that there was relatively less differences in the morphological traits between ‘Da Sajin' and ‘Xiao Sajin' than those between the former two and ‘Dan Sajin'. ‘Da Sajin' and ‘Xiao Sajin' had closer genetic relationship, but both were distantly phylogenetically related to ‘Dan Sajin'.
