摘 要:目的探讨姜黄素对非酒精性脂肪性肝病(NAFLD)大鼠环氧合酶2(COX2)表达的影响。方法 18只SD大鼠随机均分为三组:模型组和姜黄素组予以高脂高胆固醇食物10ml/kg灌胃联合四氯化碳石蜡油溶液3ml/kg腹腔注射,对照组给予生理盐水灌胃及腹腔注射,每周2次。建模成功后,姜黄素组给予姜黄素10ml/kg,对照组和模型组给予等量的羧甲基纤维素钠。治疗4周后,检测大鼠血清ALT、AST、TG和TC水平,HE染色观察大鼠肝脏组织病理学变化,免疫组化和蛋白印迹法检测大鼠肝组织COX2蛋白的表达情况。结果与对照组比较,模型组ALT、AST、TG、TC和COX2蛋白表达增高(P<0.05);与模型组比较,姜黄素组ALT、AST、TG、TC和COX2蛋白表达降低(P<0.05)。对照组大鼠肝小叶结构完整,无脂肪变性及炎症坏死情况;模型组大鼠肝小叶边界模糊,肝细胞脂肪变性,炎症坏死明显;与模型组相比,姜黄素组大鼠肝小叶结构变清晰,脂肪变性减轻。结论姜黄素可降低COX2蛋白表达,减轻NAFLD大鼠的肝脂肪变性及炎症坏死。Objective To investigate the effect of curcumin on cyclooxygenase 2(COX2)expression of rats with non-alcoholic fatty liver disease(NAFLD).Methods Eighteen SD rats were randomly divided into three groups.Group A and group B were gavaged with high fat and high cholesterol 10 ml/kg and intraperitoneally injected with carbon tetrachloride 3 ml/kg.Group C was gavaged and intraperitoneally injected with normal saline twice a week.Group A was treated with curcumin 10 ml/kg,group B and group C were treated with sodium carboxymethylcellulose.After 4 weeks,serum levels of ALT,AST,TG and TC were detected,the pathology of rat liver tissues was examined by HE staining and expression of COX2 protein was detected.Results Serum levels of ALT,AST,TG and TC and COX2 protein level were higher in group B than those in group C(P〈0.05),which were lower in group A than those in group B(P〈0.05).The structure of hepatic lobule in group C was integrated without steatosis and inflammatory necrosis.The boundary of hepatic lobule in group B was blurry and steatosis and inflammatory necrosis were obvious.The structure of hepatic lobule in group A was clear and the steatosis was alleviated.Conclusion Curcumin could decrease the protein expression of COX2 and alleviate the steatosis and inflammatory necrosis of rats with NAFLD.
