摘  要:目的探讨透明质酸(HA)与CD44对卵巢癌细胞Nanog基因表达的影响。方法应用RT-PCR、序列测定等方法检测SKOV-3细胞Nanog基因的表达。在SKOV-3细胞培养液中分别加入HA、抗CD44抗体和HA,应用Western-blot方法检测细胞及细胞核内Nanog蛋白的表达。结果 SKOV-3细胞中存在Nanog基因。SKOV-3细胞培养液中加入HA后Nanog蛋白表达增加,加入抗CD44抗体和HA后Nanog蛋白表达减少,差异均有显著意义(F=1 419.356、1 181.229,P<0.05)。结论 HA、CD44可促进卵巢癌细胞Nanog基因表达,可能在卵巢癌细胞生长、侵袭和转移过程中发挥重要作用。Objective To investigate the influence of hyaluronic acid (HA) and CD44 on Nanog gene expression in ova- rian cancer cells. Methods RT-PCR and sequencing were performed to measure the expression of Nanog gene in SKOV-3 cells. HA or anti-CD44 antibody+HA was added to the cell culture medium of SKOV-3 cells, and Western blot was used to measure the protein expression of Nanog in cells and cell nuclei. Results The Nanog gene was detected in SKOV-3 cells. There was a signifi- cant increase in the protein expression of Nanog after HA was added to the cell culture medium of SKOV-3 cells (F= 1 419.356, P〈0.05), while there was a significant reduction in the protein expression of Nanog after the addition of anti-CD44 antibody and HA (F=1 181.229 ,P〈(0.05). Conclusion HA and CD44 can promote the expression of the Nanog gene in ovarian cancer cells and thus may play an important role in the growth, invasion, and metastasis of ovarian cancer cells.
