豚鼠绝经后肾虚型原发性骨关节炎模型的建立

摘　　要：目的:探索3、5月龄Hartley豚鼠去除卵巢能否建立病证结合的绝经后肾虚型原发性骨关节炎(POA)模型。方法:将59只7周龄Hartley雌性豚鼠,抽取5只作为正常组,其余分为3、5月龄组,分别随机抽取5只作为对照组,然后随机分为去势组和假手术组。通过观察豚鼠大体状况和检测血清环磷酸腺苷(c AMP)、环磷酸鸟苷(c GMP)水平及其比值变化验证肾虚;通过连续观察阴道脱落细胞动情周期的变化和检测血清雌二醇(E2)的浓度验证绝经,通过豚鼠膝关节大体观和石蜡切片的组织学镜检及相应评分表半定量评分验证POA。结果:3、5月龄去势组都出现一系列肾虚表现,并且血清E2、c AMP降低,c GMP升高,c AMP/c GMP显著降低,且5月龄去势组更为突出;阴道脱落细胞涂片显示,3、5月龄去势组动情周期消失且E2水平显著降低;大体观及镜检显示仅5月龄去势组表现出明显的早中期OA变化,且相应的评分也支持这一结果。结论:3、5月龄去势组均可建立绝经后肾虚型POA病证结合模型,但5月龄更佳。Objective： To explore whether the removal of the ovaries in 3/5-month old Hartley guinea pigs can be used to establish a combined model of postmenopausal primary osteoarthritis （POA） and syndrome of kidney deficiency. Methods：Fifty-nine 7-week-old female Hartley guinea pigs, from which 5 pigs were extracted as the normal group, were divided into the 3/5-month group. Five guinea pigs in the two groups were randomly selected as the control groups, and the rest were randomly divided into ovariectomized （OVX） groups and sham operation （sham） groups. Through the observation of general situation and detection of the serum levels of cyclic adenosine monophosphate （cAMP）, cyclic guanosine monophosphate （cGMP） and the ratio changes of cAMP and cGMP, the model of kidney deficiency was verified. Through continuous observation of the estrous cycle changes of vaginal exfoliated cells and detection of serum concentration of estradiol （E2）, the postmenopausal model was verified. Through the gross appearances of stifles in guinea pigs and the microscopic examinations of paraffin sections and the corresponding scoring systems, the model of POA was verified. Results： The guinea pigs of OVX groups in 3 and 5 months groups had the syndrome of kidney deficiency, what was more, the serum levels of E： and cAMP decreased and cGMP increased, and the ratios of cAMP to cGMP decreased significantly. And the 5-month OVX group was more prominent. The vaginal smears showed that the normal estrous cycles disappeared in 3/5 months OVX groups, and the serum level of E2 was significantly lower. Obvious and macroscopic changes of the knees showed ihat obvious changes of OA in the early and middle stages just appeared in 5-month OVX group, which was supported by the corresponding scores. Conclusion： The removal of the ovaries in 3/5-month old Hartley guinea pigs can be used to establish a combined model of postmenopausal primary osteoarthritis and syndrome of kidney deficiency successfully.
