荷叶、葛根和鱼腥草提取物体外抗肝癌及抗HBV活性研究

摘　　要：目的：研究荷叶、葛根和鱼腥草提取物的体外抗肝癌及抗HBV活性。方法：以HepG2.2.15细胞为模型,MTT法检测三种中药对细胞增殖的影响;光学显微镜观察药物对细胞形态的影响;流式检测细胞的凋亡情况;ELISA法检测细胞上清中HBsAg和HBeAg的含量。结果：三种中药给药浓度为2000μg/m L时,对细胞的增殖具有抑制作用;三种中药浓度达1000μg/m L时,可使细胞形态发生病理性改变,显著促进Hep G2.2.15细胞的凋亡;在125-1000μg/m L浓度范围内,三种中药提取物均可显著降低细胞上清中HBsAg和HBeAg的含量。结论：荷叶、葛根和鱼腥草提取物均具有体外抗肝癌和抗HBV活性。Objective： To study the anti-hepatocarcinogenesis and anti-HBV activity of the extracts of lotus leaf,pueraria and Houttuynia cordata in vitro. Methods： The HepG2. 2. 15 cells were used as the model. MTT assay was used to detect the effect of three Chinese herbs on cell proliferation. The morphology of cells was observed by optical microscope. The apoptosis of the HepG2. 2. 15 was detected by flow cytometry. The content of HBsAg and HBeAg was measured by ELISA. Results： When the concentration of three drugs was 2000 μg/m L,they could inhibit the proliferation of the cells. When the concentration of the three drugs was more than 1000 μg/m L,drugs can cause the morphological changes and apoptosis. In the range of 125 ~ 1000 μg/m L,the contents of HBsAg and HBeAg were significantly decreased. Conclusion： The extracts of lotus leaf,pueraria and Houttuynia cordata have anti-hepatocarcinoma and anti-HBV activity in vitro.
