摘 要:目的探讨胃癌患者外周血和肿瘤组织中LIM域特有蛋白1(LMO1)、基质金属蛋白酶(MMP)9、上皮型钙黏素(E-cadherin)蛋白的表达及其临床意义。方法选取经病理学检查确诊的150例胃癌患者(研究组),以及年龄和性别相匹配的50名行健康体格检查者(对照组)。取肿瘤和癌旁组织石蜡标本经免疫组织化学染色检测肿瘤组织LMO1、MMP-9、E-cadherin蛋白的表达情况,采用ELISA法测定研究组术前、术后1个月和对照组的外周血中LMO1、MMP-9、E-cadherin蛋白表达量。结果研究组胃癌组织中LMO1、MMP-9的表达阳性率显著高于癌旁正常组织(P值均<0.01),E-cadherin表达阳性率显著低于癌旁正常组织(P<0.01)。研究组术前外周血中LMO1、MMP-9水平显著高于同组术后1个月和对照组(P值均<0.05),E-cadherin水平显著低于同组术后1个月和对照组(P值均<0.05);而研究组术后1个月上述各项指标与对照组间的差异均无统计学意义(P值均>0.05)。胃癌组织LMO1蛋白表达与淋巴结转移有关(χ~2=4.894,P<0.05)。研究组胃癌组织与外周血中LMO1表达呈正相关(r=0.351 2,P<0.001)。研究组术前外周血LMO1与MMP-9呈正相关(r=0.273 4,P<0.001),与E-cadherin呈负相关(r=-0.186 6,P=0.022)。结论监测外周血LMO1可能对反映胃癌患者的疾病状态有帮助。Objective To detect the expression of LIM-only protein 1 (LMO1), matrix metalloproteinase 9 (MMP-9) and epithelial cadherin (E-cadherin) in peripheral blood and tumorous tissues in patients with gastric cancer, and to investigate their relationship and significance. Methods A total of 150 patients with pathologically proved gastric cancer were recruited to research group, and 50 healthy persons with matched age and sex were assigned to control group. Immunohistochemical assay was performed to detect the expression of LMO1, MMP-9 and E-cadherin in gastric cancer tissues and normal mucosa, while their expression in peripheral blood was tested with enzyme linked immunosorbent assay(ELISA)before and one month after operation. The expression of LMO1, MMP-9 and E-cadherin in peripheral blood in the control group was also tested with ELISA. Results The expression of LMO1 and MMP-9 in tumorous tissues were significantly higher than those in normal mucosa (both P〈0.01), while the expression of E-cadherin was obviously lower in tumorous tissues than that in normal mucosa (P〈0.01). Before operation, the levels of LMO1 and MMP-9 in peripheral blood in the research group were significantly higher than those after operation or in the control group, while the level of E-cadherin was relatively lower in the research group than that after opteration or in the control group (all P〈0.05). But these indexes were not significantly different between the control group and the research group after operation (all P〉0.05). The expression of LMO1 protein in gastric cancer tissues was related to lymphatic metastasis (X2 = 4. 894, P〈0. 05). Significant positive correlation was found between LMO1 expression in tumor tissues and its expression in peripheral blood (r= 0. 351 2, P〈0. 01). LMO1 was positively correlated with MMP-9 (r = 0. 273 4, P〈 0. 001), and negatively correlated with E-cadherin (r = 0. 186 6, P = 0. 022) in peripheral blood in research group before operation. Concl
