摘  要:突变型Braf与多种恶性肿瘤的发生密切相关。为了研究Braf对NHE1活性的调控及对胶质瘤侵袭性的影响,本研究选用人神经胶质瘤母细胞系U251细胞,采用Matrigel-transwell方法区分侵袭能力不同的胶质瘤母细胞,通过BCECF荧光探针检测其胞内pH值,分析侵袭性不同的胶质瘤母细胞中NHE1活性的差异;并通过实时荧光定量PCR检测侵袭性不同的胶质瘤母细胞中Braf的表达水平。利用BCECF荧光探针实验和Matrigel-transwell方法检测Braf mRNA表达水平对细胞内NHE1活性及细胞侵袭能力的影响。结果表明:胶质母细胞的侵袭性受Braf表达水平的正向调控。Braf可能通过正向调控NHE1的活性,创造有利于胶质母细胞侵袭的微环境,促进细胞侵袭。The mutant Braf is closely related with variety of malignant tumors. In order to explore the regulation on NHE1 activity and the influence on invasion abilities of glioblastoma by Braf, we used matrigel-transwell to separate strong-invasive and weak-invasive subpopulations of U251 cells. Then the NHE1 activity in strong and weak-invasive subpopulations ofglioblastoma was analyzed when BCECF measured the pH. The expression levels of Braf in glioblasts with different invasiveness were detected by RT-qPCR. BCECF fluorescene probe assay and Matrigel-transwell method were used to detect the effect of Braf mRNA expression level on the activity of NHE 1 and the invasiveness of cells. The results showed that the invasion abilities of glioblastoma were positively regulated by the Braf expression level. Ideal Braf might positively regulate the activity of NHE1 to create a microenvironment conductive to glioblast invasion and promote cell invasion.
