摘 要:目的:脂肪酸合成酶(fatty acid synthase,FASN)是脂肪酸(fatty acid,FA)合成代谢的关键酶,在肿瘤的发生、发展过程中扮演了重要角色。本研究拟探讨FASN在膀胱癌组织中的表达及其临床意义。方法:运用Oncomine数据库挖掘FASN相关的基因芯片信息,分析FASN在膀胱癌组织中的表达及其与膀胱癌类型和患者预后的关系;收集2012年1月~2016年12月我院收治的膀胱癌患者的膀胱癌组织50例和对应的正常膀胱组织10例,采用免疫组化法检测组织标本中FASN的表达,分析其表达与膀胱癌临床病理特征的关系。结果:Oncomine数据库芯片数据显示,FASN在膀胱癌组织中的表达明显高于正常膀胱组织(P<0.05或P<0.001);FASN的表达量与患者总体生存率呈负相关(P=0.0162)。免疫组化显示:FASN在膀胱癌组织中较正常膀胱组织高表达,分别为89.21%和43.57%(P<0.01),在浸润性癌组织中较非浸润性癌组织高表达(P<0.05),其表达水平与膀胱癌临床分期、病理分级相关(P<0.05)。结论:FASN在膀胱癌组织中高表达,与膀胱癌临床病理特征密切相关,与患者预后呈负相关,它可能是膀胱癌潜在的调控因子,或可为膀胱癌的早期诊断、预后评估及靶向分子治疗提供新的切入点。Objective: Fatty acid synthase (FASN) is a key enzyme of fatty acid biosynthesis metabolism and plays an important role in the occurrence and development of tumors. The purpose of this study was to investigate the expression and its clinical significance of FASN in bladder carcinoma. Method: Oncomine database was used to extract the useful mining gene chip database for analyzing the expression of FASN in bladder carcinoma tissues and its relationship with the tumour type and the prognosis of patients. Totally 50 cases of bladder cancer tissues and 10 cases of normal bladder tissues were collected during the period from January, 2012 to December, 2016 in the department of urology of our hospital. The expression of FASN in tissues was detected by immunohistochemistry, and the relationship between the expression and clinicopathologic characteristic of bladder cancer was analyzed. Result: Oncomine database showed that the expression of FASN in bladder cancer tissues was significantly higher than that in normal bladder tissues (P〈0.05 or P〈0. 001), and it was negatively correlated with the overall sur- vival rate of bladder cancer patients (P=0. 0162). The results of immunohistochemistry showed that FASN was overexpressed in bladder cancer tissues (89.21N) compared with the normal bladder tissues (43.57%, P〈0.01), and higher in invasive carcinoma tissues than that in non-invasive carcinoma tissues (P〈0.05). Also, its expression was correlated with the clinical stage and pathological grade of bladder cancer (P〈0.05). Conclusion: FASN is highly expressed in bladder cancer, and closely related to the clinicopathologic characteristic of bladder cancer, and negatively correlated with the prognosis of patients. It is suggested that FASN might be a potential regulator of bladder carcinoma, or may provide a new breakthrough point for the early diagnosis, prognosis evalu- ation and targeted molecular theranv for bladder carcinoma.
