摘 要:目的:探讨miR-186与EGFR在宫颈癌组织和细胞中的表达及作用机制。方法:RT-PCR与Western blot法分别检测30例宫颈癌组织与30例正常宫颈组织中miR-186与EGFR表达,分析其与宫颈癌临床病理之间的关系。增殖侵袭实验检测SiHa和HeLa细胞增殖与侵袭;双荧光素酶实验明确EGFR的3'-非翻译区(3'-UTR)是否为miR-186的直接靶点。结果:与正常宫颈组织比较,宫颈癌组织中miR-186呈低表达,EGFR呈高表达,两者均与宫颈癌分化程度、淋巴转移密切相关。宫颈癌组织中miR-186表达与EGFR表达均呈负相关(EGFR mRNA:R^2=0.865,P<0.001;EGFR蛋白:R^2=0.832,P<0.001)。上调miR-186表达能抑制SiHa和HeLa细胞的增殖与侵袭。双荧光素酶实验明确miR-186靶定了EGFR的3'UTR并影响了EGFR的蛋白表达。EGFR过表达逆转了miR-186抑制的细胞增殖和侵袭。结论:miR-186通过靶定EGFR的3'-UTR抑制EGFR表达进而抑制宫颈癌SiHa和HeLa细胞的增殖与侵袭。Objective:To investigate the expression of miR-186 in cervical cancer tissues and cells,and to discuss its mechanism implicated in the development of cervical cancer.Methods:The expression levels of miR-186 and EGFR were detected by RT-PCR and Western blot in 30 samples of cervical cancer tissue and 30 samples of normal cervical tissues.The relationships between miR-186 expression and clinicopathological features of cervical cancer were analyzed.CCK-8 assay was used to analyze the effects of miR-186 on the proliferation and invasion of cervical cancer cells SiHa and HeLa.Luciferase assays were used to determined whether EGFR-3'UTR was the direct target of miR-186.Results:miR-186 expression in the cervical cancer tissues was significantly lower than that in the normal cervical tissues,while the expression of EGFR was higher than that in the normal cervical tissues. The expression miR-186 was inversely related to the expression of EGFR( EGFR mRNA:R^2= 0. 865,P〈0. 001;EGFR protein:R^2= 0. 832,P〈0.001).The expression level of miR-186 in the cervical cancer tissues was significantly correlated with degree of differentiation and lymphatic metastasis up-regulated miR-186 expression would significantly reduce the proliferation and invasion of SiHa and HeLa. Luciferase assays showed that miR-186 binds the EGFR-3'UTR and affect the expression of EGFR protein.EGFR over-expression reversed miR-186-inhibited cell proliferation and invasion.Conclusions:miR-186 expression can inhibit the proliferation and invasion of cervical cancer Si Ha and He La cells through targeting EGFR-3'UTR.
