摘  要:目的:探讨钙黏附蛋白11(cadherin-11,CDH11)甲基化在宫颈癌中的表达及临床意义。方法:应用甲基化特异性聚合酶链反应检测CDH11基因在3株宫颈癌细胞系、1株人表皮永生化细胞株、30例宫颈癌组织、17例正常宫颈组织中的甲基化状态。统计患者临床资料,分析甲基化同临床病理特征之间的联系。结果:CDH11在3株宫颈癌细胞系中,均出现不同程度甲基化;在1株人表皮永生化细胞株中未检测到甲基化。CDH11在宫颈癌组织中的甲基化率为56.7%(17/30),显著高于正常宫颈组织23.5%(4/17),差异具有统计学意义(P<0.05)。CDH11基因甲基化在不同分期的宫颈癌组织中检出率无统计学意义(P>0.05);在不同年龄段、是否有淋巴结转移以及分化级别上,CDH11基因的甲基化率差异无统计学意义(P>0.05)。结论:宫颈癌组织中CDH11基因甲基化水平高于正常宫颈组织,差异有统计学意义。CDH11基因甲基化可能是宫颈癌独特的基因甲基化谱成员之一,可以成为宫颈癌早期诊断的新的标志物。Objective:To investigate the expression and clinical significance of CDH11 gene methylation in cervical cancer.Methods:CDH11 methylation was detected by methylation specific PCR(MSP)in 3 cervical cancer cell lines,1 normal human immortalized keratinocytes cell line,30 cases of cervical cancer fresh specimens and 17 normal cervical tissues.Patients' clinicopathological features were collected and analyzed.Results:MSP revealed that CDH11 gene methylation founded in 3 cervical cancer cell lines,and in 56.7% of cervical cancer patients 4 of 17(23.5%)normal cervical tissues showed methylation.No methylation was detected in HacaT cell line and normal cervical tissues.The methylation of CDH11 was not associated with age,FIGO stage,lymph node metastesis and classification.Conclusion:The CDH11 promoter methylation which occurs in cervical cancer was significantly higher than the normal controls.Our results indicated that CDH11 gene methylation as a frequently event involving in the tumorigenesis of cervical cancer,and may be expected to be a new marker to predict the occurrence of patients with cervical cancer.
