摘　　要：目的:探讨LNK基因在急性白血病患者中的表达及其与临床特征的相关性。方法:采用实时荧光定量PCR检测80例AL(ALL 42例,AM L 38例)患者骨髓中LNK m RNA的表达水平,分析其表达水平与临床特征的相关性,应用Western blot检测患者的LNK蛋白表达量。以正常的志愿者(16例)骨髓作为对照组。结果:ALL和AM L组的LNK水平均明显高于正常对照组(P=0.007,P=0.021),ALL组与AM L组之间LNK水平无统计学差异;ALL和AML组中的LNK表达水平均与危险程度呈正相关(P=0.000,P=0.004,r=0.5,r=0.386),且ALL和AML组中高危组的LNK水平均明显高于对照组(P=0.035,P=0.032),AML和ALL组的中危组(P=0.239,P=0.609)及标危组(P=0.974,P=1)LNK水平均高于对照组,但无统计学意义,各组之间危险程度无统计学差异。急性白血病患者LNK蛋白表达量较对照组增加。结论:急性白血病患者LNK基因表达高于正常人,与患者危险度呈正相关。Objective： To investigate the expression of LNK gene in patients with acute leukemia （AL） and its correlation with the clinical characteristics. Methods： Real-time quantitative RT-PCR was used to detect the level of LNK mRNA in bone marrow samples from 80 patients diagnosed as AL（42 cases of ALL, and 38 cases of AML）, and its relevance with clinical indicators was statistically analyzed. Western blot was used to detect the expression of LNK protein. The bone marrow samples of 16 healthy volunteers were used as the controls. Results： The LNK mRNA levels in ALL and AML groups were significantly higher than that in control group （P =0.007, P =0.021 ） and there was no statistical difference between ALL and AML groups. The LNK levels in ALL and AML groups possitively correlated with the risk of patients （ P = 0. 000, P = 0. 04, r = 0.5, r = 0. 386 ）, And the LNK levels in high risk ALL and AML groups were significantly higher than that in control group （ P = 0. 035, P = 0. 032）, the LNK levels in intermediate risk of AML and ALL groups （ P = 0. 239, P = 0. 609 ） and the LNK level in standard risk （ P = 0.974, P = 1 ） were all higher than that in control group, there was no statistianl significance, but the risks of different groups showed no statistical significance. The LNK protein level in patients with acute leukemia was higher than that in control group. Conclusion： The expression level of LNK gene in AL patients is higher than that in healthy people, and the expression level of LNK gene positively correlates with the risk of patients.
