摘  要:目的通过检测癫痫患者外周血液血清细胞因子白介素1β(IL-1β)、白细胞介素2(IL-2)、白细胞介素6(IL-6)、白细胞介素8(IL-8)和肿瘤坏死因子α(TNF-α)水平变化,探讨癫痫患者发病机制与免疫学的相关性。方法选取2014年6月一2016年6月期间于北京中医药大学孙思邈医院就诊的癫痫患者100例和同期体检健康人群100名。采用酶联免疫吸附法(ELISA)分别对血清细胞因子IL-1β、IL-2、IL-6、IL-8以及TNF-α含量水平进行测定,应用SPSS21.0软件进行统计学分析。结果癫痫患者血清中IL-1β、IL-2、IL-6、IL-8、TNF-α的含量水平显著高于正常对照组人群(P<0.01)。在100例癫痫病人中,按年龄(≤35岁与>35岁)、性别(男性组与女性组)以及病程(≤1年较短病程组与>1年较长病程组)分层分析表明,血清细胞因子IL-1β、IL-2、IL-6、IL-8和TNF-α的含量水平与患者年龄、性别、病程长短没有相关性(P>0.05)。Pearson直线相关性分析表明,癫痫患者血清细胞因子IL-1β、IL-2、IL-6、IL-8的表达量分别与TNF-α的含量水平呈正相关变化(P<0.001)。结论细胞因子含量水平的失衡参与了癫痫发生发展的免疫病理过程,IL-1β、IL-2、IL-6、IL-8以及TNF-α可能参与了癫痫发病的免疫学机制。Objective To investigate the changes of serum interleukin-1β( IL-1β),interleukin-2( IL-2),interleukin-2( IL-6),interleukin-2( IL-8) and tumor necrosis factor-alpha( TNF-α) levels in epileptic patients,and to analyze their correlations with the pathogenesis of epilepsy.Methods A total of 100 eligible epileptic patients( epilepsy group) and another 100 healthy people( control group) were recruited at our hospital between June 2014 and June 2016. Serum content of IL-1β、IL-2、IL-6、IL-8 and TNF-α of the two groups was detected with ELISA. The data was analyzed with statistical software SPSS 21.0.Results Compared with control group,serum concentrations of IL-1β、IL-2、IL-6、IL-8 and TNF-αwerer significantly increased( P〈0.05).The levels of IL-1β、IL-2、IL-6、IL-8 and TNF-α were not significantly different between patients of different age,sex and course of disease in epilepsy group( P〉0.05). Levels of serum IL-1β、IL-2、IL-6 and IL-8 were positively correlated with TNF-α in epileptic patients( P〈0.05).Conclusion The immune system of patients with epilepsy is in an activated state. It is possible that the cytokines IL-1β、IL-2、IL-6、IL-8 and TNF-αmay be involved in the pathogenesis of epilepsy.
