摘  要:目的:通过针刺足三里穴,观察癫痫大鼠海马组织中细胞因子Leptin、RANTES的表达,以探讨针刺治疗癫痫的作用机制。方法:选用6~8周龄体质量约200~250 g的SD雄性大鼠30只,随机分为正常空白组、正常足三里组、模型空白组、模型足三里组。采用氯化锂-匹罗卡品癫痫大鼠模型,于造模成功后第2天予以针刺干预,连续10天后取材,运用液相芯片技术检测各组大鼠海马组织中Leptin、RANTES的表达。结果:与正常空白组相比,模型空白组RANTES的表达明显增高,差异有统计学意义(P〈0.05),而Leptin的表达虽有所增加,但差异无统计数意义(P〉0.05),正常足三里组Leptin、RANTES的表达均有所下降,而差异无统计学意义(P〉0.05);与模型空白相比,模型足三里组Leptin、RANTES的表达均有所下降,且差异有统计学意义(P〈0.05);正常足三里组与模型足三里相比,RANTES的表达增加,差异有统计学意义(P〈0.05),而Leptin的表达下降,差异无统计学意义(P〈0.05)。结论:针刺足三里穴可通过降低癫痫大鼠海马组织Leptin、RANTES的表达,促进癫痫大鼠机能的恢复,起到抑制癫痫的作用;癫痫状态下,针刺足三里能够促进大鼠海马组织RANTES的特异性表达。Objective: The expression of cytokines Leptin and RANTES in hippocampus of epilepsy rats was observed by acupuncture at Zusanli( ST36) to explore the mechanism of acupuncture in the treatment of epilepsy. Methods: 6 ~ 8 weeks old male SD rats weighing about 200-250 g were randomly divided into normal control group,normal Zusanli( ST36) group,model control group,model Zusanli( ST36) group. Lithium chloride-pilocarpine rat model of epilepsy was set up. At the second day after modeling,we used acupuncture intervention,consecutively for 10. And then we used liquid chip technology to detect the expressions of Leptin and RANTES in hippocampal tissue of rats. Results: Compared with normal blank group,the expressions of Leptin and RANTES in normal Zusanli( ST36) group decreased,but the difference was not statistically significant( P 〉 0. 05). RANTES expression in the blank model group was significantly increased and the difference was statistically significant( P 〈 0. 05),while the expression of Leptin increased,but there was no statistical significance( P 〉 0. 05). Compared with the model blank,the expressions of Leptin and RANTES in the Zusanli( ST36) model group was decreased and the difference was statistically significant( P〈 0. 05). Compared with normal Zusanli( ST36) group,the expressions of RANTES in the Zusanli( ST36) model group was increased and the difference was statistically significant( P 〈 0. 05),while the expression of Leptin decreased and the difference was not statistically significant( P 〈 0. 05). Conclusion: Acupuncture at Zusanli( ST36) can reduce the expressions of Leptin and RANTES in hippocampus of epilepsy rats and promote the recovery and inhibit epilepsy. Under the status of epilepsy,acupuncture at Zusanli( ST36) can promote the specific expression of RANTES in rat hippocampus.
