摘 要:目的通过构建糖尿病兔、梅毒兔及糖尿病合并梅毒兔模型并进行基因芯片检测,对比各组实验兔肾脏脂肪沉积情况和筛选与之有关的脂肪代谢异常基因。方法耳缘静脉注射四氧嘧啶诱导构建糖尿病兔(DM组)、睾丸接种梅毒螺旋体构建梅毒兔(Tp组)和四氧嘧啶构建糖尿病模型后接种梅毒螺旋体构建糖尿病合并梅毒兔(H组)模型,分别在建模成功后第2、4、6、8、10、12和14周,抽取耳缘静脉血检测尿素氮(BUN)与肌酐(Cr),14周后处死实验兔,取各组兔肾组织进行油红O染色、HE染色了解肾脏脂肪变损伤情况及进行基因芯片检测,筛选脂肪代谢有显著差异的基因。结果成功构建了糖尿病兔(DM组)、梅毒兔(Tp组)和糖尿病合并梅毒兔(H组)模型;各组实验兔均发生肾功能损害且随病程的进展越来越严重,与其它组同期相比H组肾损害更明显;油红O染色发现H组肾组织脂滴沉积最密集;基因芯片检测发现ACOX1、ACADM、SCP2基因在各组实验兔脂肪代谢中差异有统计学意义,与正常组相比,3个基因在Tp、DM、H组表达均下调(P<0.05),H组下调最为显著(P<0.05)。结论糖尿病合并梅毒组(H组)兔肾脏损伤和脂肪沉积最为严重;实验兔ACOX1、ACADM、SCP2基因表达下调可能与肾脏脂肪沉积相关。Objective The fat deposition of kidneys of rabbits in each group was compared and the abnormal genes related to fat metabolism were screened by constructing the rabbit models of diabetes mellitus, syphilis and diabetes mellitus combined with syphilis. Methods The rabbit models of diabetes mellitus (the DM group) were established by injecting alloxan in ear vein, the rabbit models of syphilis (the Tp group) by vaccinating Treponema pallidum (T. pallidum) in testis, and the rabbit models of diabetes mellitus combined with syphilis (the H group) by injecting alloxan before vaccinating T. pallidum. Nitrogen (BUN) and creatinine(Cr) were detected by ear vein blood at the 2nd, 4th, 6th, 8th, 10th and 14th weeks respectively. After 14 weeks, fatty deposits of kidneys of the rabbits in each group were detecteded by oil red O staining and HE staining. Abnormal adipose metabolism gene candidates in renal tissues were screened by gene chip technique. Results The diabetes mellitus rabbit models, the syphilis rabbit models and the diabetes mellitus combined with syphilis models were constructed successfully. Renal function damage was found in the rabbits of each experimental group, and it was getting worse as the disease progression. Compared with other groups, the H group showed the severest renal function damage. The Results from oil red O staining showed that lipid droplet deposition in renal tissues of the H group was the most intensive. The Results from gene chip detection indicated that ACOX1, ACADM and SCP2 genes in adipose metabolism of rabbits in experimental groups showed statistically significant differences. The expression of the above-mentioned three genes in the groups of Tp, DM and H was down-regulated as compared with the normal group, and the most significant down-regulation was found in the H group. Conclusions The rabbit models of diabetes mellitus combined with syphilis (the H group) show the severest renal damage and fat deposition. The down-regulation of ACOX1, ACADM and SCP2
