摘 要:目的探讨血液系统疾病染色体异常的临床意义。方法观察16例血液系统疾病患者的细胞遗传学核型及骨髓细胞形态与疾病之间的关系。在无菌条件下抽取骨髓标本2~3ml,肝素抗凝,加入到RPMI1640培养基中,采用短期培养法制片,分析20~30?个中期分裂细胞,以G显带进行染色体核型分析。结果 16例血液病患者均有染色体核型异常,异常克隆包括结构异常和数目异常。其中结构变异型染色体的改变较为少见,如急性髓系白血病有t(1;2)t(8;21)和慢性粒细胞白血病有ins(1;5)t(9;22);数目异常以+8,+21,-Y等易见。结论 16例血液系统疾病患者存在染色体异常,可见染色体核型分析在血液系统疾病诊断、分型、预后判断中有着重要的临床价值。Objective:To discuss the clinical significance of chromosome abnormality in hematological diseases. Methods Analyze the relation between cytogenetic karyotype and cellular morphosis of bone marrow in 16 patients with hematological diseases. Extract 2-3 ml sample of bone marrow with heparin anticoagulant under sterilized conditions,add to the RPMI1640 medium,short-term culture and chromosome preparation,chromosomal abnormalities were analyzed 20-30 metaphase cells by G-banding. Results:16 patients with hematological diseases had the chromosomal abnormalities,aberrant clones with chromosome abnormalities contain Structural Abnormality and numerical changes. Chromosomal structural abnormality was rare,acute myeloid leukemia t(1;2)t(8;21) and chronic myeloid leukemia ins(1;5)t(9;22);numerical changes +8,+21,-Y. Conclusion:16 patients with hematological diseases had the chromosomal abnormalities,Chromosome karyotype analysis showed significant clinical value in diagnosis,classification and prognosis of hematological diseases.
