宫颈癌组织中Bcl-2相关抗凋亡基因3表达及意义

摘　　要：目的探讨Bcl-2相关抗凋亡基因3(BAG3)在宫颈癌组织中的表达情况及临床意义。方法分别采用免疫组织化学法和实时荧光定量PCR技术,检测103例宫颈癌(宫颈癌组)、50例子宫颈上皮内瘤变(CIN组)和40例正常宫颈(对照组)组织中BAG3蛋白和BAG3基因的表达情况。利用Kaplan-Meier生存曲线分析BAG3蛋白表达对宫颈癌病人生存时间的影响。结果宫颈癌组BAG3蛋白阳性率均高于CIN组和对照组,差异有统计学意义(χ2=27.167、28.514,P<0.01);宫颈癌组BAG3mRNA相对表达量均高于CIN组和对照组,差异有统计学意义(t=30.453、26.295,P<0.01);宫颈癌组BAG3蛋白和mRNA表达量均与FIGO分期、分化程度、淋巴结转移和肌层浸润有关(χ~2=4.691~7.530,P<0.05;t=28.825~35.696,P<0.01);Kaplan-Meier生存分析显示,BAG3蛋白阳性表达组平均生存时间为37.96个月,BAG3蛋白阴性表达组则为53.70个月,Log-Rank检验差异有统计学意义(χ2=4.555,P<0.05)。结论 BAG3在宫颈癌组织中呈高表达,可能参与了宫颈癌的发生发展和侵袭转移过程,并与病人的预后密切相关。因此,BAG3有望成为宫颈癌诊断及预后判断的潜在生物学标志物。Objective To examine the expression of Bcl-2-associated athanogene 3（BAG3）in cervical cancer and its clinical significance.Methods The BAG3 gene and BAG3 protein expression in 103 cervical cancer tissues,50 cervical intraepithelial neoplasia（CIN）tissues,and 40 normal cervical tissues were measured by immunohistochemistry and quantitative real-time PCR.The effect of BAG3 protein expression on the survival of cervical cancer patients was analyzed by the Kaplan-Meier survival curve and log-rank test.Results The BAG3 protein expression rate was significantly higher in the cervical cancer group than in the CIN and control groups（χ^2=27.167 and 28.514,both P〈0.01）.Similarly,the relative BAG3 mRNA expression was significantly higher in in the cervical cancer group than in the CIN and control groups（t=30.453 and 26.295,both P〈0.01）.BAG3 protein and mRNA expression in cervical cancer tissues was correlated with FIGO staging,differentiation,lymph node metastasis,and myometrial invasion（χ^2=4.691-7.530,P〈0.05;t=28.825-35.696,P〈0.01）.Kaplan-Meier survival analysis showed that the mean survival time was significantly shorter in the BAG3 protein-positive group than in the BAG3 protein-negative group（37.96 months vs 53.70 months,χ^2=4.555,P〈0.05）.Conclusion BAG3 is highly expressed in cervical cancer tissues.It may be involved in the development,progression,invasion,and metastasis of cervical cancer,and is closely associated with patient prognosis.Therefore,BAG3 is a potential biomarker for cervical cancer diagnosis and prognosis.
