摘 要:目的观察雌激素干预C57BL/6小鼠大部分胰腺切除后胰岛功能的变化,探讨雌激素对胰岛β细胞的影响。方法 60只小鼠随机分成4组:对照假手术组(Sham)、胰腺切除模型组(Px)、雌激素干预组(E2)和雌激素阻断剂组(E2+ICI),每组15只。ELISA检测血浆胰岛素水平;小鼠处死前进行葡萄糖耐量实验(GTT);处死后荧光染色胰岛,计算胰岛数量和体积,并进行葡萄糖刺激胰岛素分泌实验(GSIS);RT-PCR检测Ins-1、Ins-2和PDX-1基因的表达水平。结果胰腺切除手术后7 d,E2组血糖较Px组下降[(162±18)vs(225±11)mg/dl),P<0.05]。GTT结果显示,服糖120 min后,E2组与Px组比较,血糖回落到基线水平[(145.5±24.5)vs(235.0±21.3)mg/dl,P<0.05]。胰岛荧光染色结果表明,各手术组的胰岛体积明显增大(P<0.05),尤其是E2组胰岛体积增大最为明显。GSIS结果发现,雌激素能显著增加胰岛素释放及胰岛素含量。RT-PCR结果提示,雌激素能增加胰岛Ins-2基因的表达。结论雌激素干预可降低糖尿病小鼠血糖水平,增加胰岛体积和胰岛素分泌,并对胰岛素基因的表达产生影响。Objective To investigate the effect of 17β-estradiol on islet β-cell function in C57 mice after partial pancreatectomy. Methods 60 mice were randomly divided into 4 groups: control sham operation group ( Sham), pancreatectomy model group (Px), estrogen intervention group ( E2 ) and estrogen blocker group (E2+ICI) ,15 in each group. Glucose tolerance test (GTT) and glucose-stimulated insulin secretion assay (GSIS) were performed before sacrifice. The islets were stained by fluorescence after the mice were sacrificed. The count and volume of islet were calculated. Plasma insulin levels were measured by ELISA. The expressions of Ins-l, Ins-2 and PDX-1 gene were detected by RT-PCR. Results The blood glucose level in E2 group was significantly lower than that in Px group at 7 days after operation [(162±18) vs (225±11) mg/dl,P〈0. 05]. Compared with Px group,the blood glucose of E2 group dropped to baseline level at 120 min after glucose Stimulation [(145.0±24. 5) vs (235.0±21.3) mg/dl, P 〈 0.05] .The islet volume of each operation group markedly increased after partial pancreatectomy (P〈0. 05), especially in E2 group. The insulin content and the expression of Ins-2 gene were increased in E2 group. Conclusion E2 intervention can reduce the blood glucose, increase the islet volume and insulin secretion and affect the expression of insulin gene in Px mice.
