摘  要:目的研究竹节参皂苷保护D-半乳糖致SH-SY5Y神经细胞损伤的作用及机制。方法采用50g/L的D-半乳糖刺激SH-SY5Y神经细胞,以不同浓度竹节参皂苷(1,5,25μg/mL)和50g/L的D-半乳糖共同孵育SHSY5Y神经细胞24h。JC-1试剂盒检测细胞线粒体膜电位,Western blot检测细胞Sirt1蛋白的表达。结果竹节参皂苷能显著升高线粒体膜电位和Sirt1蛋白的表达。结论竹节参皂苷通过激活Sirt1抑制线粒体功能障碍从而改善D-半乳糖对SH-SY5Y神经细胞造成的损伤。Objective To observe the protective effect and mechanism of Panax japonicus saponins on D-galactose induced SH-SY5 Yneuron injury.Methods The 50 g/L D-galactose was used to stimulate SH-SY5 Yneuron,Different concentrations Panax japonicus saponins(1,5,25μg/mL)and D-galactose co-incubated with SH-SY5 Yneuron for24 h.The changes of mitochondrial membrane potential were identified by JC-1 detection.Protein level of silentmating type information regulation 2 Homolog-1(Sirt1)was detected by Western blot analysis.Results Panax japonicus saponins significantly improved mitochondrial transmembrane potential and the expression of Sirt1 protein.Conclusion Panax japonicus saponins inhibits mitochondrial dysfunction by activating Sirt1,thereby improving D-galactose induced damage to SH-SY5 Yneuron.
