摘  要:目的:在Cuff法("袖套"法血管吻合术)小鼠颈部异位心脏移植术的基础上,改良出一种效果更稳定、个体差异更小、术程更简易的小鼠颈部异位心脏移植模型。方法:制备供体时直接阻断下腔静脉并同时4℃肝素灌注,后立即剥离供心置于冰盘上冷却停跳。在移植过程中使用套管法吻合肺动脉与颈外静脉,主动脉与颈总动脉,将心脏总缺血时间降低到8min以内。结果:模型较以往方法术程明显缩短,手术难度降低,移植物热缺血时间<1min,并使每次手术间冷缺血时间变得稳定可控,续重复C57BL/6J小鼠同系移植20对,24h存活率为97%,14d后移植物窦性跳动的18对,成功率为90%,大大高于缝线法颈部移植和腹部移植术式。结论:本文介绍的模型较传统模型更为简单、快速、稳定、可靠,可以广泛推广。Objective: On the basis of the cuff’s (“sleeve cuff” method of vascular anastomosis) cardiac heterotopictransplantation in neck wi th mouse, we developed a new model of mouse to mouse heterotopic by modified cuf f technique, tt has a more stable e f fe c t, smaller individual differences and easier operation process. Methods:The donor. postcavawas perfused by the 4℃ donor heparin immediately thereafter d ire c t ly occluded, the donorwas stripped and placed on an ice tra y to cardioplegia. In the process of transplantat ion, the lar vein,the aorta and the common carotid a r te ryw e re fitted by cuff ’ s cannulation to reduce the the heatt to wi thin 8 minutes. Results:Compared wi th the conventional method, the operation time of the modll was sig-nificantly shortened, the operation di f f icul ty was reduced, the warm ischemia time of the g ra t t was less and the time of cold ischemia in each operation became stable and controllable. In the 20 isografts by C57B L /6J mice,97% donor heart still beats after 24 h o u rs,in 14 days, 8 donors were beating heart sinus,the succesignificantly higher than stylol it ic neck transplantation and abdominal transplantation. Conclusion: T he model presentedin this paper is simpler, faster,stable and rel iable than the trad itional model. It can be widely spread.
